Surface plasmon resonance sensing is a new technology. This method has many advantages, such as high stability and anti-electromagnetic interference and testing without damaging, etc. So it is widely used in biochemical, medical diagnostics, life sciences , environment pollution monitor and so other areas. With the wide intersection and application of different fields of science, such as informatics, material science, mechanism, and microelectronics etc, SPR biosensor technology is getting matured gradually and created a new commercial market, which has a good prospect. After theoretical research and model analysis to SPR phenomenon, an multi-layer SPR biosensor system is designed and made for laboratory experiments. The main research work is shown as following:1. Principle and exciting condition of SPR phenomenon is discussed detailedly;2. ZnO thin films with excellent optical characteristics is prepared on the bottom of prism by PLD technology. The crystal characteristics are detected by X-ray diffraction and scanning electric microscopy technology.3. The optical constant of thin films is measured by m-line method, which provides experimental parameters for the design of multi-layer SPR biosensor.4. Three media with different dielectric (air, pure water and dirty water) is tested. Through analyzing the ATR curves, we can conclude: the multi-layer SPR biosensor exhibits a more sharp-angled ATR curves and a great sensitivity enhancement.
