Experimental purposes:The aim of the experiment is that discussion on the relationship between apoptosis of germ cell of high-fat diet-induced testicular and inflammatory factors in obese rats CRP, MCP-1, and also Initially revealed that aerobic exercise intervention on dietary fat male spermatogenic cell apoptosis and inflammation in rats.Methods:The healthy male SD rats were randomly divided into four groups:Normal Diet Group(C), High-Fat Diet Group(HF), Regular Diet Exercise Group(CE), High-Fat Diet Exercise Group(HE), n=12, total 48; group C and group CE was given normal diet,group HF and group HE was given high-fat, high-calorie diet. After the experiment, the number of rats in each group: 8 in Group C,11 in group HF,10 in Group CE, and 11 in group HE. Exercise program was 60min-No swimming motion for 10 weeks, six days every week. After the end of the experiment, serum triglycerides(TG), total cholesterol(TC), high density lipoprotein(HDL), low density lipoprotein of rats were detected(LDL); of body weight, body length, epididymal fat weight, Lee’s index were measured and calculated body fat ratio;C-reactive protein(CRP), monocyte chemoattractant protein 1 (MCP-1) levels were determined with ELISA and Western blot methods; testicular in CRP and MCP-1 expression positions in tissue in immunohistochemistry were assayed; CRP, MCP-1 expression in testis and epididymal fat weight was detected by RT-PCR; changes in testicular tissue morphology was observed by HE staining and rat testis cell apoptosis was detected in situ end labeling port.Results:1. After 10 weeks, compared with group C, rats’ weight in CE was significantly reduced, and Lee’s index, TG decreased significantly, while significantly increased HDL and there is no significant difference in terms of testis week fat, body fat ratio, TC, LDL. HF group, compared with group C, body weight, testis week fat, body fat ratio, Lee’s index, LDL, TC showed a significant increase, TG, HDL were not significantly different. Group HE, compared with group HF, body weight, epididymal fat weight, body fat ratio, Lee’s index, also TC, TG, LDL were significantly reduced, HDL was no significant difference.2. Spermatogenic cells in group C seminiferous tubules developed normally,tightly arrangement, clear cell structure, a large number of mature spermatozoa in the lumen, and can clearly see the testicular seminiferous tubules between quality cells; seminiferous tubule wall were thinning in the HF group, with spermatogenic cells arrangement in disorder, the number of layers of spermatogenic cells and mature sperm number significantly reducing,visible cell shedding phenomenon, and seminiferous tubule basement membrane was incomplete, obvious reduce of Leydig cells; in group CE, Testicular seminiferous tubules of rats was normal, and Arranged basically in order of spermatogenic cells and increase in cell gap can be seen; group HE spermatogenic cells developed normally, and cells shedding phenomenon occasionally, and also seminiferous small tube interstitial tissue distributed loosely3.Compared with the group C. serum CRP levels in HF group was significantly increased, as well as MCP-1 levels. Group HE, compared with group HF, CRP, MCP-1 levels were significantly reduced. Analysis and conclude that high-fat diet can lead to CRP and MCP-1 levels in serum increase, while aerobic exercise can reduce serum CRP and MCP-1 level.4. Compared with group C, epididymal fat weight in CRP, MCP-lmRNA levels increased in the group CE, but no significant difference, while the CRPmRNA level increased significantly, MCP-1mRNA levels in HF group wereelevated significantly. HE group, compared with HF group, CRPmRNA levels reduced dramatically, MCP-1mRNA levels were decreased markedly. Thus, it can be seen that high fat diet leads to fat rat testis weeks in CRP, MCP-1mRNA elevated levels of testosterone cause inflammation of epididymal fat weight, while aerobic exercise can reduce CRP and MCP-1 mRNA levels appropriately, and reduce inflammation state.5. Compared with group C. HF group CRP, MCP-1mRNA levels significantly increased:group HE. compared with group HF, CRP and MCP-1mRNA levels significantly reduced. It can be seen that high-fat diet contribute to rats testis CRP, levels of MCP-1mRNA are elevated, and lead to testicular inflammation, while aerobic exercise can reduce CRP, MCP-1 mRNA levels, reduce inflammation state.6. Protein expression assayed by Western blot, it can be seen that group HE, compared with group C. CRP, MCP-1 protein levels were significantly increased; group HE, compared with group HF, CRP, MCP-1 protein expression levels were significantly reduced, and testicular inflammatory caused by high-fat diet can be reduced through exercise.7. Immunohistochemistry results:CRP, MCP-1 expression cannot be seen in the testes of rats in group C; in high-fat diet group HF, CRP expression can be found in Leydig cells, and MCP-1 in Leydig cells and Sertoli cells had expression partly; in group CE and group HE, CRP levels expression in rat testis tissue was low relatively, and MCP-1 in HE group was also observed rarely while the there was no MCP-1 expression CE group found. Rats aerobic exercise can reduce CRP, MCP-1 expression levels.8. The situation of apoptosis in rat testis:Compared with group C, apoptosis rate of testis germ cell in group CE was increased, but no significant difference. HF group and HE group testis germ cell apoptosis rate significantly increased. Group HE, compared with group HF, testis germ cell apoptosis rate decreased significantly. It found that high-fat diet may cause testicular tissue inflammation, leading to an increase in testis germ cell apoptosis, while aerobic exercise can aerobic exercise can ease the situation.Conclusion:1. The high-fat diet can lead to male SD rats lipid metabolism disorders, and aerobic exercise can improve this situation.2. The high-fat diet can make SD male rat testis tissue structure change, and enables the spermatogenic cell apoptosis to increase significantly, the situation can be improved by aerobic exercise.3. There is close relationship between apoptosis of rats’sperm production and the expression of CRP.MCP-1, whereas CRP, MCP-1 expression and sperm apoptosis rate of rats who did aerobic exercise also decreased, suggesting that the body may lead to spermatogenic cell apoptosis when the high-fat diet-induced inflammation happens, reducing inflammatory factors:CRP, MCP-1 expression through aerobic exercise to intervene germ cell apoptosis in rats who has obesity inflammation.
