Objective:To establish ulcerative colitis dampness-heat intestinal animal model, study its pathogenesis and explore the mechanism of supplemented pulsatilla decoction in treating dampness-heat intestinal ulcerative colitis model rats.Material and method:48 SPF wistar rats were selected with about 160-180g. The diet and drinking method+ DDS+acetic acid hypoclysis were adopted for further modeling. The rats were divided into four groups randomly including control group, model group, west medicine group (mesalazine) and Chinese medicine group (supplemented pulsatilla decoction) (12 rats per group). The rats in the control group were fed normally. The rest of three groups were fed by high-sucrose-fat diet with 10ml/kg 56% liquor for 10 days. The rats started to drink 5% DSS solution by themselves in the 11th day for 7 days. The 8% acetic acid solution was used to conduct coloclysis after 24h with fasting food but not water in the 17th day. After successful modeling, the mesalazine and supplemented pulsatilla decoction were used to conduct coloclysis on the western medicine group and Chinese medicine group respectively for two weeks, during which the experiment observed the rats’ general conditions. Then, the rats were killed as experiment materials. Besides, this experiment conducted pathological detection to determine the changes of P-selectin and ICAM-1 as well as IFN-y and IL-4 expression in the colon tissues.Results:1.Compared with control group, the rats in the rest of groups had diarrhea with bloody mucopurulent stool, laziness, gathering together, lusterless hair and decreasing weight etc at the first day of modeling. Further, the rats in the western medicine group and Chinese medicine group had normal stool, improved spirit, increased activity amount, slow luster hair, increased water consumption and weight recovery with about one-week drug use.2.Compared with control group, the histopathological score (HS) in the model group got improved obvisouly (P<0.05). The HS in the western group and Chinese medicine group was obviously lower than that in the model group (P<.0.05).3.The IFN-y in the rats’ colon tissues of model group was obviously higher than that in the rats of control group (P<0.01); the supplemented pulsatilla decoction and mesalazine can reduce the IFN-y in the rats’colon tissues of model group; the IL-4 in the rats’ colon tissues of model group was obviously lower than that in the rats of control group (P<0.01); the supplemented pulsatilla decoction and mesalazine can increase the IL-4 in the rats’colon tissues of model group.4.The P-selectin in the rat serum of model group was obviously higher than that in the rats of control group (P<0.01); the supplemented pulsatilla decoction and mesalazine can decrease the P-selectin in the rat serum significantly; the ICAM-1 in the rat serum of model group was higher than that in the rats of control group (P<0.01); the supplemented pulsatilla decoction and mesalazine can reduce ICAM-s.Conclusion:1. The high-sucrose-fat diet and drinking method+DSS+acetic acid hypoclysis can make the dampness-heat intestinal ulcerative colitis model.2. The pathogenesis of dampness-heat intestinal UC was the rising IFN-y in the colonic mucosa; the decreasing IL-4 would cause Th1/Th2 imbalance; the rising ICAM-1 and P-selectin in the serum promoted rats’ inflammatory response.3. The supplemented pulsatilla decoction had obvious therapeutic effects on dampness-heat intestinal UC.
