Objective1.To investigate the expression of FXR、SREBP-lc and FAS in nonalcoholic fatty steatosis hepatocyte models induced by oleic acid.2.T0 investigate the effects of berberine on the expression levels of FXR、SREBP-lc and FAS in the nonalcoholic fatty liver disease cell model induced by oleic acid.MethodsUpon oleic acid inducing HepG2 cell, non-alcoholic fatty liver disease cell model was established. Oil red O staining and intracellular triglycerides were detected for observing the situation of intracellular hepatocyte steatosis. The HepG2 cells were divided into control group、oleic acid-treated group, Which the control group cultured in DMEM medium, NAFLD group were treated with lmmol/L oleic acid treated for 24 h. The expression levels of FXR、SREBP-lc and FAS mRNA and protein were measured by RT-PCR、 Western-blotting.HepG2 cells were divided into control group, NAFLD group, berberine-treated group (low-dose、middle-dose、high-dose).Oil red O staining and intracellular triglycerides were detected for observing the situation of intracellular hepatocyte steatosis.The expression of FXR、SREBP-lc and FAS mRNA and protein were measured by RT-PCR、Western-blotting.Results1.The HepG2 steatosis model was established successfully with the treatment of lmmol/L oleic acid.Oil red O staining showed that the contents of intracellular lipid droplets increased significantly, and the levels of intracellular TG increased to 262.8(ug/mg protein) from 127.3(ug/mg protein)(P<0.01). Compared with the control group, the expression levels of FXR mRNA and proteins significantly decreased in the oleic acid-treated group(P<0.01);SREBP-lc and FAS mRNA and proteins significantly increased in the oleic acid-treated group(P<0.01).2. After treatment with different dose of berberine for 24 hours, oil red O staining showed that the contents of intracellular lipid droplets decreased, and the levels of intracellular TG decreased (P< 0.05). The expression levels of FXR、SREBP-1c and FAS and proteins reverted to the levels of the control group after treatment with different dose of berberine.ConclusionThe expression of hepatocyte FXR in nonalcoholic fatty steatosis models were decreased, however, SREBP-lc and FAS were increased, which may provide experimental basis for the further treatment of some non-alcoholic fatty liver disease children.Berberine can improve liver steatosis,improve the expression levels of FXR in the nonalcoholic fatty liver disease cell model, decrease the expression of SREBP-lc and FAS, which indicate that berberine may improve liver steatosis through FXR/SREBP-lc/FAS signaling pathways.
