Mechanism Of Liver Parenchymal Injury Induced By Chronic Stress - Induced Depression In Rats And The Protective Effect

Research purposesTo investigate the effect and mechanism of chronic stress on liver parenchyma and function.Based on this,we want to further observe the effect of traditional Chinese medicine compound Sini Powder intervention on stress-induced liver injury.Research methodsChronic stress models were prepared by Mild Unpredictable Stress chronic(CUMS)and chronic restraint stress in rats.We passed the test scores of rats in the open field and sucrose preference degree to evaluate whether depression in rats.We can move through the detection of enzyme alanine aminotransferase in rats(ALT)and aspartate aminotransferase(AST)content, to observe the rat liver function injury.At the same time,we performed pathological examination of liver biopsy,observe whether rat liver cell damage,in order to determine the degree of liver injury in rats.Through the detection of free fatty acids in rat liver(FFA),triglyceride(TG),total cholesterol(TC)content of oil red O staining,we can observe the lipid metabolism of rats.Then we detected rat liver tumor necrosis factor(TNF-α)and interleukin 6(IL-6),superoxide dismutase(SOD)and malondialdehyde(MDA)content.So we can study the mechanism of the effect of chronic stress on the two main pathways(lipid metabolism and oxidative stress).Research results1.Cums and chronic restraint stress can lead to a decline in the overall situation of the organism of rats and mice.Kuang game scoring and sucrose preference degree decreased significantly.Rats showed a "depressed" state.The stress depression model of rats was successfully reproduced by two kinds of chronic stress.2.The contents of ALT and AST were significantly increased in two kinds of stress depression model rats,which suggested that stress induced depression resulted in the injury of liver function.Rat liver HE staining showed that the liver cells had edema,increased volume,liver disorder,inflammatory cell infiltration and so on.Therefore,CUMS and chronic restraint stress induced depression can damage the liver function of rats,and cause injury of liver in rats.3.In the chronic restraint stress model,the serum FFA content of rats was significantly increased,the content of TG and TC in serum and liver was significantly increased,and the content of inflammatory factors TNF-a and IL-6 were increased.Oil red staining was also confirmed by the lipid droplets which we observed in the hepatic cell vacuoles.The result indicated that the chronic restraint induced stress induced depression can cause the abnormal lipid metabolism and the disorder of lipid metabolism in rat liver,which leads to the accumulation of fat in the liver cells.4.In the chronic restraint stress model,the SOD activity in the liver of rats was decreased,and the content of MDA increased.Therefore,chronic restraint induced stress induced depression can lead to liver tissue lipid peroxidation damage.5.Traditional Chinese medicine compound Sini powder can significantly improve the depression symptoms of two kinds of stress depression model in rats.Its antidepressant effect was similar to that of fluoxetine hydrochloride.Four inverse scattered can significantly reduce the content of ALT,AST,and gradually repair the damage of liver function.Sini powder can significantly reduce the serum levels of TNF-a and FFA,TG,TC,IL-6 content.Therefore,Sini scattered can gradually correct the disorder of lipid metabolism in rats,reduce the inflammatory reaction and lipid peroxidation damage in rats.Research conclusion1.CUMS stress and chronic restraint stress can induce the depression of SD rats,and then cause the liver function and liver cell damage.2.CUMS stress and chronic restraint stress may be caused by liver lipid metabolism and oxidative stress injury caused by liver injury.3.Sini powder can significantly improve the depressive symptoms of rats,and has the effect of anti depression.Sini powder energy in the treatment of liver function damage,fatty liver disease,chronic inflammation has a significant therapeutic effect.And it has the trend of treatment for the abnormal lipid metabolism and oxidative stress.Therefore,Sini powder has the role of anti liver injury.
