Aims: In this project we attempted to use a kind of optical material, Up-Converting Phosphor (UCP), in the immunoassay as the reporter. And by making fully use of its optical properties, a UPT-based biosensor can be devised to detect many kinds of pathogen and their corresponding antibody quantitatively.Methods: After systematical treatments of the surface of standardized UCP particles, make UCP particles conjugate with many kinds of biological molecule covalently. Then by screening the materials of immunochromatographic strip on the large scale, develop a kind of new strip using UCP particles as reporter. Finally take advantages of the optical properties of UCP particles to devise a portable biosensor to detect the pathogen and the antibody quantitatively.Results: [1] By controlling the concentration and quantity of reagents and the speed and time of the reaction, we got the optimized method to produce standardized UCP particles. [2] After silica-coating, amine-functionalizing and aldehyde-functionalizing, UCP particles with activated surface could conjugate with a variety of biological molecules stably. And during this period in order to ensure the quality of these treatments, we developed a series of QC methods. [3] Using UCP particles that conjugated with antibody to combine with immunochromatographic assay, we developed the UPT-based strip that could detect the antibody against pestis with relatively high sensitivity and specificity. [4] Based on the optical properties of UCP particles and the UPT-based strip, we devised the UPT-based biosensor that could reveal the precise concentration of detected substance in the sample. [5] Eventually, by combing the technique of UCP production, UCP surface modification and activation, UPT-based immunochromatographic strip and UPT-based biosensor together, we got the complete UPT system and established the standard method of quantitative detection. Conclusion: To be tested by practical application, the UPT system has already realized the quantitative detection and had the primary ability to meet the need of some emergencies.
