Objective: Study CXCR7 protein expression in serous epithelial ovarian cancer cell line SKOV3 and mucinous epithelial ovarian cancer cell line 3AO, investigate the difference of CXCR7 protein expression and proliferation of ovarian cancer before and after cisplatin (DDP), Inferred the relationship between CXCR7 and development of ovarian cancer.Methods:(1) Using immunocytochemical staining CXCR7 expression in ovarian cancer cell line SKOV3, 3AO. (2)Using MTT assay study proliferation ability. Of SKOV3, 3AO cells after cisplatin (3)Using western blot detection CXCR7 expression of epithelial ovarian cancer cell SKOV3, 3AO before and after cisplatin (DDP).Results: (1) CXCR7 protein expression were positive in SKOV3, 3AO cells, CXCR7 protein expressed in the cytoplasm,. Cancer cells structure were clear, dyeing of CXCR7 expression cells is significantly higher than background under microscope,; (2) Using MTT assay stain proliferation of overain cancer cells, results: there were no significant difference under the concentration of 5ug/ml(DDP) compared with the control group. Difference between concentrations of 10ug/ml, 20ug/ml, 40ug/ml groups and the control group,concentration 5ug/ml group were significantly different. There were significant difference between concentration of 10ug/ml group and concentration of 20ug/ml, 40ug/ml groups;Comparing datas of 24h reaction time group and 48h, 72h reaction time group,there were significantly different between two groups. (3) western blot results: CXCR7 expression between SKOV3 group and 3AO group was no significant difference .There were significantly different between cisplatin intervention SKOV3, 3AO cell groups and control groups, CXCR7 expression in cisplatin intervention SKOV3, 3AO cell groups was significantly inhibited.Conclusions: (1) chemokine receptor CXCR7 were positive expressed in serous epithelial ovarian cancer cell line SKOV3 and mucinous epithelial ovarian cancer cell line 3AO ,there were no significantly different between two ovarian cancer groups. (2) the minimum concentration effective concentration of cisplatin is 10ug/ml, when reaction time was 24h, the proliferation of two groups ovarian cancer cells were significantly reduced; CXCR7 protein were expressed in the two ovarian cell lines after cisplatin intervention, But its expression were significantly reduced...
