AIMS:This topic through the establishment of type 1 diabetic mice and type 2 diabetic rat model,observed the role Cortex Mori flavonoids on diabetic animals,thus provide experimental data on treatmenting diabetes.METHODS:(1) Extract Cortex Mori flavonoids by ultrasonic wave and drawn the standard curve by rutin,then determin the content.(2) KM male mice intraperitoneal injection with STZ(140mg/kg), selected blood glucose greater than 14.2mmol/L to type 1 diabetes successful model .The successful model mice were randonmly divided into Cortex Mori flavonoids high, medium , low dose group and model group,n=5. Set 5 as the normal control group . Blood glucose levels and hepatic glycogen content were measured after administration 10 days.(3) Wistar male rats were injected intraperitoneal with STZ(40mg/kg) after feeding with high sugar and fat diet for 4 weeks. Take another 5 rats injected with citrate buffer solution ,then feed with high sugar and fat diet together. Fasting blood glucose greater than 14.2mmol/L were selected for the type 2 diabetic rats.The diabetic rats were randonmly divided into Cortex Mori flavonoids high, medium , low dose group, Rosiglitazone control group and model group,n=5.Select 5 citrate buffer injected rats for normal control group. Fasting blood glucose levels,triglyceride and hepatic glycogen content were measured after administration 14 days. RESULTS:(1) The content of Cortex Mori flavonoids was 23.1% after ultrasonic extraction method.(2) Compared with model group, Cortex Mori flavonoids high dose group reduced blood glucose levels (P <0.05), hepatic glycogen content increased (P <0.05), Cortex Mori flavonoids medium dose and low dose group no significant difference.(3) Diabetic rats after 2 weeks of administration, compared with the model group, Cortex Mori flavonoids high dose group and the medium dose group were significantly lower blood glucose (P <0.01, P <0.05), low dose group were not significantly different. Cortex Mori flavonoids decrease the levels of blood glucose in a manner of dose-dependence; Triglyceride levels, Cortex Mori flavonoids high dose group than in model group was significantly lower (p <0.05), medium dose group and low dose group no significant difference; Compared with model group , hepatic glycogen content in Cortex Mori flavonoids high dose group were significantly higher, significantly different (P <0.05), medium dose group and low dose group have no significant difference.CONCLUSIONS:(1) Ultrasonic extraction method is high efficiency,low loss and short extraction time.(2) High dose Cortex Mori flavonoids can low blood glucose level of the type 1 diabetic mice ,increase hepatic glycogen conten.Cortex Mori flavonoids can low blood glucose and triglyceride level of the type 2 diabetic rats,increase hepitac glycogen content. It is suggused that Cortex Mori flavonoids has effect of anti- Diabetes mellitus.
