| Objective:To observe the effects of Tetramethylpyrazine(TMP) on proliferation, apoptosis and cell cycle of hepatic stellate cell-T6 (HSC- T6), so as to explore Tetramethylpyrazine's antihepatic fibrosis effects and its nechamism.Methods:HSC-T6 cell strain was cultured in vitro, and was effected with varying doses(10mg/l,50mg/l,100mg/l,200mg/l,400mg/l,600mg/l)of TMP and 0.005mg/l[1] of transforming growth factor-β1(TGF-β1). MTT chromatometry was applied to measure the HSC-T6 proliferation inhibition ratio, flow cytometry to measure the cell apoptosis and the percentage of DNA content in different group.Results:Compared with blank group, all the TMP groups had a obviously higher proliferation inhibition rate and apoptosis rate, is significant difference(P<0.01). As the TMP concentration wsa increased, prolifer- ation inhibition rate and apoptosis rate of HSC-T6 to increased gradually. Compared with blank control group, cell percentage in G0/G1 phase was obviously increased and was obviously degraded in S phase in all TMP groups. As to add TMP concentration, the cell percentage was gradually increased in G0/G1 phase and gradually decreased in S+G2 phase.Conclusion:TMP can inhibit the proliferation and induce apoptosis in concentra- tion dependent way of HSC-T6. The mechanism of TMP can inhibit the prociferation of HSC-T6 possibly due to making HSC-T6's cell cycle arrest in G0/G1 phase, prevent HSC-T6 to ingress S phase. |
