Objective: To investigate effects of sinomenine on the kidney pathology and Bcl-2/Bax in kidney tubular epithelium of Sprague-Dawley Rats with Ischemia-Reperfusion Injury. Discuss the kidney ischemia-reperfusion injury effective prevention and treatment methods. For the clinical application of sinomenine prevention kidney ischemia-reperfusion injury provided the experiment evidence and theoretical basis.Methods: Fifty four Sprague Dawley rats were randomly divided into nine groups, each group of six rats: (1) Ischemia reperfusion control group;(2)IRI group 0h;(3) IRI group 24h; (4) IRI group 48h; (5) IRI group 72h; (6) sinomenine treatment IRI group 0h;(7) sinomenine treatment IRI group 24h; (8) sinomenine treatment IRI group 48h; (9) sinomenine treatment IRI group 72h. The levels of serum creatinine and urea nitrogen were measured; Renal histology injury was assessed by histological damage scores; Expression of Bcl-2 and Bax was detected by immunohistochemistry.Results: (1)The levels of serum creatinine and urea nitrogen in model group were obviously elevated, and significantly different from those in control group (P<0.05). In treatment group, the levels of serum creatinine and urea nitrogen were significantly lower compared with model group(P<0.05), while higher than those of control group (P<0.05); (2)The histological damage scores in model group signific- antly different from those in control group and treatment group(P<0.05), while higher than those of control group (P<0.05); (3)Compared with the results of control group in which the expressions of Bcl-2 and Bax on renal tubules were slight, the expressions of Bcl-2 and Bax boosted in model group (P<0.05); The treatment group showed a significantly high expression of Bcl-2(P<0.01), along with decreased expression of Bax(P<0.05).Conclusion: SIN can attenuate SCr and BUN values, and renal tubule impairments after renal ischemia-reperfusion injury in rats, which indicates a protective effect renal ischemia-reperfusion injury.The mechanism may be associated with inhibition of apoptosis affect the expressions of Bcl-2 and Bax.
