| Objective To study the relationship among the polymorphism rs13266634C/T of SLC30A8 gene and T2DM and IGR in the Hengyang Population of China.Methods According to the diagnostic criteria of WHO in 1999, a total of 574 unrelated Hengyang people was randomly selected, 236 T2DM(males,121, females, 115; average age, 57.4±10.5),120 IGR(impaired glucose regulation, males, 57; females, 63; average age, 53.9±11.5)and 218 NGT(without T2DM family history, males, 109; females, 109; average age, 54.2±1.1) were recruited in the second affiliated Hospital of Nan Hua University. 5ml blood was taken from the elbow vein of each subject. Human genomic DNA was extracted by chloroform method. Clinical characteristics such as BMI, BP, TG, TC, HDL-C, LDL-C, etc, were measured and calculated meanwhile. Genotyping for the SLC30A8 gene rs13266634 polymorphisms was carried out by PCR-RFLP analysis(polymerase chain reaction-restriction fragment length polymorphism), HOMA-β, HOMA-IR were analyzed to estimateβ-cell function and insulin resistance.Results1. BMI, SBP, DBP, LDL-C, FPG levels of type 2 diabetic patients were higher than the patients with IGR(P<0.05 or 0.01), Patients with type 2 diabetes had higher Age, BMI, SBP, DBP, LDL-C, TC, TG and FPG level than those of normal controls(P<0.05 or 0.01), the patients with IGR had higher BMI, SBP, DBP, LDL-C, TC than those of NGT(P<0.05 or 0.01.2. In T2DM group, the frequencies of C allele were significantly higher than that in NGT group(χ2=22.73,P=0.000), the frequencies of C allele of the IGR group were also significantly higher than that in NGT group(χ2=7.77, P=0.003). CC genotypes frequencies of rs13266634C/T in the T2DM group and IGR group were significantly higher than that in NGT group. The C allele of rs13266634C/T significantly increased T2DM and IGR risk with an allelic odd ratio(OR) of 1.89 and 1.57.3. HOMA-βlevel of the patients who take with CC gene was significantly lower than CT and TT gene, and the levels in CC, CT, TT showed a growing trend.Conclusions1. SLC30A8 rs13266634C/T genetic polymorphism is associated with T2DM and IGR, and C allele is probably the risk gene of T2DM.2. SLC30A8 gene may increase the infectlousness of T2DM through affecting the function of islet cell. |
