Backgroud:Rubescensin(Oridonin,rubescensineB)posses remarkable pharmacological action, especially its anticancer potential. It is likely to be used as an chemopreventive and therapeutic drug in the future. However, the effect of Oridonin on A549 cells anti-proliferation is unclear.Objective:To investigate the effect of oridonin on A549 cells anti-proliferation.Methods:A549 cells were respectively treated with different concentration of Oridonin of 10μg/ml,20μg/ml,40μg/ml,60μg/ml for 24hrs. The proliferation of A549 cells was evaluated by means of cell counting and MTT assay, and the distribution of A549 cells cycles was detected by Flow Cytometry(FCM). and then the protein amount of Caveolin-1,and caspase-3 was analyzed by Weston Blot.Results:Oridonin could inhibitA549 cell proliferation and promote apoptosis, with the peak in 40μg/ml. MTT test observation, cells from the control group of 1.6±0.16×106/ml down to 40μg/ml oridonin treated group 0.6±0.08×106/ml; Cell flow technique observations, PI values from the control group decreased to 0.28±0.023 to 40μg/ml oridonin treated group 0.08±0.006,40μg/ml Oridonin could inhibit A549 cells from G0/G1 phase to phase S/G2/M. AO/EB staining, with the increasing concentration of oridonin, A549 gradually increased the number of apoptosis, the cell size smaller, capsule shrinkage, nuclear enrichment, chromatin aggregation, untreated A549 cells, large and round, less cytoplasm, nuclear large, some cells can be seen one to several nucleoli. Western blot detection, oridonin concentration-dependent increase in A549 cells, caveolin-1 and caspase-3 expression, when the concentration of 40μg/ml oridonin when, Caveolin-1 and caspase-3 expression in the largest quantity, Caveolin-1 gray scan values from the blank group 0.5±0.045 up to1.8±0.001, when40μg/ml. Caspase-3 gray-scale scan values from the control group of 0.3±0.005 up to 1.5±0.06when 40μg/ml. Interestingly, oridonin obviously activation caspase-3 expression, which appeared a Positive correlation with oridonin stimulated caveolin-1 activation. Conclusion:Oridonin induced A549 cells apoptosis possibly via the caveolin-1/caspase-3 pathway.
