Objective:The establishment of rat VT models and them with electro-acupuncture (EA) at Ximen (PC 4)。To observed those VT rats heart rate change and the lasting time for arrhythmia and we detected the contents of NO and activity of NOS in the rats blood plasma respectively in this way, we are trying to explore the mechanism of cupuncture therapy in VT。Methods:70 SD rats were randomly divided into four groups:normal grouop (n=10), Model group,EA at Ximen group and EA at Chize group (20 rats in each group)。Rats in model group, EA at Ximen group and EA at Chize group were made ventricular tachycardia models with cesium chloride (CsCl) injected into femoral vein。EA at Ximen group and EA at Chize group were respectively treated by EA at Ximen and Chize for 5 minutes after the rats were made ventricular tachycardia models。Each 10 of Model Group,EA at Ximen group and Chize group, heart rate change and the lasting time for arrhythmia were observed. Then, we detected the NO contents (with nitrate reductase method) and activity of NOS (with colorimetry) in the rats' blood plasma respectively。Results:1. Compared with the former and latter of Electrocardiogram, after CsCI injected inio femoral vein.we found that Model group,EA at Ximen group and EA at Chize group heart rate from Sinus rhythm to ventricular arrhythmia, Model group and EA at Chize heart rate changed little, after VT model had been built successfully for 6min, but EA at Ximen group heart rate reduced greatly。Compared with instant after model being made successfully, There were obvious difference (P<0.01).There were obvious difference between the two groups in heart rate(P<0.01)。2. Compared with the duration of VT and the total duration of arhythmia with the rats in Model group,EA at Ximen group and EA at Chize group, the duration of EA at Ximen group was abbreviated obviously than Model group (P<0.01), but EA at Chize was abbreviated than Model group (P<0.05)。3.we detected the NO contents (with nitrate reductase method) and activity of NOS (with colorimetry) were increased obviously(p<0.01) in the rats'blood plasma by EA on Ximen group,on the contrary the NO contents and activity of NOS were decreased greatly (P<0.01); EA on Ximen group were better than EA on Chizen group (P<0.05)。Conclusion:1. The therapy of electro-acupunture(EA) at Ximen acupoint obviously decreased the ventricular tachycardia rats heart rate, but the of effect of electro-acupunture (EA) at Chize acupoint is not as Ximen acupoint. indicating that electro-acupunture can antagonize arhythmia and the role of acupoints have relative specificity。2. The content of NO and activity of NOS decrease markedly after CsCI injected inio femoral vein to VT model。3. Compared with the Model group and EA at Chize group, the content of NO and activity of NOS in EA at Ximen group in the rats plasma increased obviously, and approximately approached to the normal level, indicating that EA at Ximen acupoint can markedly regulate the content of NO and activity of NOS in the rats plasma, its mechanism maybe that EA at Ximen can restrain heart sympathetic nerve and exhilarat vagus nerve so as to regulate rat's heart rate。4.
