| Objective:To study Uncaria and its extracts to the effects of blood pressure from normal animals and hypertensive model animals, screening effective area of Uncaria with blood pressure lowering through the combination of to pharmacological experiment and high performance liquid chromatography. By observing the various Uncaria extracts with antihypertensive effect on experimental endocrine hypertension animals and selecting a number of experimental indicators of different levels to seach for the mechanism, it's important to provide the theoretical basis that the prevention and treatment of hypertension and invention of new compounds for blood pressure lowering.Methods:â… Take Wistar rats and KM mice as study object, by subcutaneous injection of testosterone propionate, copying and comparing the animal model of hypertension. Dose of testosterone propionate:rat dose of 5mg/kg, mouse dose of 0.22mg/kg, for 14 days. Observe animal behavior and body weight changes in general and the animals in each group were measured blood pressure and heart rate, serum TC and TG indicators.â…¡Extract Uncaria herbs with different concentrations of methanol, the following separation to the ethyl acetate extract, butanol and water site location. Separation of the ethyl acetate extract, a total of 20 streams were obtained. Fr 3 and Fr 7 fractions were further purified which resulted of 6 crystalline finally, analysis of components from Uncaria extracts by HPLC.â…¢Hypertensive rats were randomly divided into 10 groups. The positive group received 2.16mg·kg-1·d-1 of captopril suspension and Uncaria dose in each experimental group were 90g (crude drug)/kg. Model group was given for the same amount of distilled water for 12 days. Each group were measured systolic blood pressure and heart rate before and 24h after administration 30,60,120,240,480 min. During the experiment, while observing the general behavior of each group, take measure of body weight, blood pressure and heart rate values before and first 3 days,6 days,9 days and 12 days after the administration of in each group.IV Through tail vein, take the blood to measure blood serum TC, TG, HDL-C, ALT, AST, SOD, MDA, CRE, BUN level and determine of organ coefficient.V The normal mice were given the extract of Uncaria. After 18h fasting, the mice were fed blank glucose solution and extract of Uncaria, respectively, with 90g/Kg of Uncaria extract crude drugs and 2g/Kg glucose dose. Through tail vein, take the blood to measure blood glucose value, with the intragastric administration before (Omin) and 30,60,120 min after.Results:â… Testosterone can cause unusual animal behavior generally, slow weight gain. After 2 weeks of modeling, blood pressure of model group mice was up (30.2±7.88mmHg), the heart rate also increasing, while levels of serum TC and TG was significantly higher than the control group (P<0.01).â…¡Uncaria 45,90 mg dose group were significantly blood pressure lowering in normal mice (P<0.05), especially for the 90mg dose group. Uncaria 90 mg group, after 120min, mice can significantly reduce heart rate (P <0.05). Uncaria extract group, ethyl acetate extract group after 12 days treatment, the mice improved mental state, fight less, hair gradually restore the luster. Body weight of mice were weighed, the treatment group and hypertension group, there was no significant difference (P>0.05). Uncaria extract, ethyl acetate, from 1 to 12 days after administration, endocrine hypertension mice significantly reduced the role of SBP and HR slowing, its antihypertensive effect with the increase of frequency of administration.â…¢Long-term administration, the Uncaria extract can continue to lower blood pressure and improve the general behavior and body weight abnormalities caused by modeling. The level of serum indicators also improved significantly. It is resulted that the ethyl acetate extract of Uncaria parts could reduce the serum levels of malondialdehyde (MDA) level and improve superoxide dismutase (SOD) activity.Conclusion:â… It is feasible for setting up hypertension model by subcutaneous injection of testosterone propionate, which is simple for the economic utility and good for reproducibility and stability.â…¡Uncaria and its extracts, have obvious hypotensive effect both on hypertension model mice and normal mice, characterized by the first step-down, after the boost, then after a continuous blood pressure lowering, moderate and lasting antihypertensive effects and presented in a certain range of drug dose-effect relationship. At the same time, the role of heart rate has slowed down.â…¢Long-term administration, the Uncaria extract group, ethyl acetate extract can continue to lower blood pressure, and improve the general behavior and body weight abnormalities caused by modeling.â…£Uncaria extract significantly reduced of the serum TC, TG and increased HDL-c increase the level caused by endocrine modeling and to some extent, reduced the free radicals to inhibit peroxidation. |
