Objective:Colorectal cancer is one of the most common malignant tumor.In the digestive tract tumors,the incidence of colorectal cancer after gastric cancer and esophageal cancer,and its incidence and mortality were tested tended to increase year by year.Many studies have shown that the incidence of colorectal cancer is a result of the joint effect of multiple genes. This study by detecting tumor suppressor gene PTEN(phosphatase and tensin homology deleted on chromosome 10),integrin- linked kinase (ILK),CD34 (microvessel density, marked to reflect the level of activity of the angiogenesis-specific antibodies) protein in colorectal cancer tissues,adjacent tissues and normal colorectal tissue expression,combined with clinical and pathological data analysis to explore tumor suppressor gene PTEN,ILKand CD34 expression in colorectal cancer development, invasion and metastasis in an attempt to provide a theoretical treatment of colorectal cancer based on.Methods:1 65 cases of colorectal carcinoma, the corresponding adjacent tissues and normal colorectal tissues samples were collected at the Fourth Hospital of Hebei Medical University 2th department of Surgery in November 2008- June 2009. All the tissues were fixed in 4% paraformaldehyde,conventional dehydration and embedded in paraffin.2 The expression of PTEN protein, ILK and CD34 were detect by S-P immunohistochemical stain(IHC) in 65 colorectal carcinoma,adjacent carcinoma and normal colorectal tissues.Results:1 The expression results of PTEN protein,ILK,and CD34(microvessel density, MVD) value in colorectal cancer tissues,adjacent tissues and normal colorectal tissuePTEN protein in colorectal cancer tissues, adjacent tissues and normal colorectal tissues positive rates were 32.31% (21/65), 50.77% (33/65) and 100% (65/65),the expression of PTEN protein in cancer group was significantly lower than the adjacent group and normal group,the difference was significant(P<0.05),the adjacent group and normal group no significant differences(P> 0.05).ILK in colorectal carcinoma tissues,adjacent tissues and normal colorectal tissues positive rates were 72.31% (47/65), 33.85% (22/65), 27.69%(18/65),in the cancer group The expression of ILK was significantly higher than the adjacent group and normal group,the difference was significant(P <0.05),the adjacent group and the the normal group was no significant difference(P>0.05),MVD values in the cancer group,the adjacent group and normal group was 35.21±10.24,12.93±5.43 and 6.55±0.88,the cancer group significantly higher than the adjacent cancer group and normal group,the difference was significant(P<0.05),adjacent group was higher than the normal group, the difference was significant(P <0.05).2 The relationship between the PTEN protein,ILK,MVD value with the pathological features and staging of colorectal cancer Expression of PTEN protein,ILK and MVD value weren't correlated with age,sex,tumor location,tumor size,tumor general sub-typing and tumor tissues in general sub-typing and histological type was no significant difference (P>0.05),but correlated with pathological grade, the depth of the tumor invaded the bowel wall, condition of the lymph node metastases, Dukes stage and distant place metastasis(P<0.05).3 The relationship of PTEN and ILK65 cases of colorectal carcinoma, PTEN protein expression in 21 cases of positive and negative in 44 cases, ILK expression in 47 patients with positive and negative in 18 cases. Expression of PTEN protein and ILK positive in 9 cases, were tested negative in 6 cases; PTEN protein expression in 44 cases negative, ILK, 38 were positive, ILK expression in 18 cases negative, PTEN protein, 12 cases were positive. The Spearman rank correlation analysis, with a negative correlation between the expression of (r=-0.510, P<0.05).4 The correlation between MVD value and PTEN,ILKThe MVD value(38.82±8.89)in colorectal carcinoma with positive PTEN protein expression was significantly lower than that with negative PTEN protein expression(23.05±7.80), there were stastically significance (t=6.774, P<0.05).And with the expression degree increasing, MVD value also increased, correlation analysis showed that MVD value had a negatively correlation with PTEN (r=-0.559, P<0.05).The MVD value(39.90±8.48) in colorectal carcinoma with positive ILK expression was significantly higher than that with negative ILK expression (24.34±7.91), there were stastically significance(t=7.400, P<0.05). And with the expression degree increasing, MVD value also increased, correlation analysis showed that MVD value had a positive correlation with ILK (r=0.574, P<0.05).Conclusion:1 PTEN protein in colorectal cancer tissues was significantly lower than the adjacent tissues and normal colorectal organization, suggesting that the absence of PTEN may be related to the occurrence of colorectal cancer; PTEN protein expression and tumor pathological grade, depth of wall invasion, lymph node transfer, Dukes stage and distant metastasis, suggesting that low expression of PTEN protein in colorectal cancer invasion and metastasis.2 ILK in colorectal cancer tissues was significantly higher than the adjacent tissues and normal colorectal organization, suggesting that ILK may be related to the development of colorectal cancer; ILK expression and tumor pathology in patients with grade, depth of wall invasion, lymph node metastasis, Dukes stage and distant metastasis, suggesting that high expression of ILK colorectal cancer invasion and metastasis.3 MVD values in the expression of colorectal cancer was significantly higher than the adjacent tissues and normal colorectal organization, MVD values in patients with pathological tumor grade, depth of wall invasion, lymph node metastasis, Dukes stage, distant metastasis and vascular tumors in bolt significantly correlated, suggesting that MVD is a measure of the value of colorectal cancer development, invasion and metastasis of important indicators.4 The low expression of PTEN protein and ILK,CD34 value in colorectal carcinoma was consistent, suggesting that low PTEN expression, ILK high expression in colorectal cancer may be related to the development process of angiogenesis is closely related; and PTEN and ILK has a Correlation.
