Objective:On the base of its bacteriostasis parsts, we investigate the multi-effects of Sargentodoxa cuneata on peritoneal adhesions in rats by vitro bacteriostasis test,vivo animals experiment and cytokine determination, in order to approach the mechanism of Sargentodoxa cuneata effective parts on peritoneal adhesions in rats.Methods:(1)Sargentodoxa cμneata is extracted with 70% ethanol in 75℃heat,then to analysis the content of chlorogenic acid and saponin in sargentodoxa cuneata by using High Performance Liqμid Chromatography (HPLC) and spectrophotometry, after the ethanol extracts was separated by the macroporous adsorptive resins.(2)To evaluate the bacteriostatic action by comparing the MIC, using 2-fold agar dilution method to statistic MIC about Escherichia coli,Klebsiella pneμmonia, Pseμdomonas aerμginosa, and Enterococcus faecalis of standard and clinical strains.(3)Rat model of peritoneal adhesions was established with cecal ligation plus puncture (CLP), and to observe the temperature and weight, to view blood flow in microcirculation of intestinal tissues by LDF, Moreover, adhesive tissues were obtained for detection of hydroxyproline by spectrophotometry and TNF-a, TGF-β1, MMP-1,TIMP-1,VEGF and ICAM-1 by the method of ELISA.Results:The content of chlorogenic acid and saponin are 0.17% and 0.13%. Both elixation and the effective part of Sargentodoxa cuneata have some inhibition effect to all experimment stains, especially to Enterococcus faecalis,the effective part has stronger bacteriostatic action than the elixation.The rat model of peritoneal adhesions was established successfully,easily and stablized.The action site can increase blood flow in microcirculation of intestinal tissues in some degree.Treated with the action site,the contents of OHP was lower than others.At the same time, TNF-α, TGF-β1, TIMP-1,VEGF and ICAM-1 were decreased,but MMP-1 was increased.Conclusions: The chlorogenic acid and saponin in Sargentodoxa cuneata have defenitely bacteriostasis effects. To verify the multi-effects of chlorogenic acid and saponin on bacterial peritonitis from the whole and cytokine level after making sure the bacteriostasis sites,that can be prevent peritoneal adhesions effectively. |
