The expression of CyclinA, p21WAF1 and C-myc proteins were detected by the immunohistochemical method of S-P. And the expression of CyclinA mRNA, p21WAF1 mRNA were detected by in situ hybridization. The expression (optical density and positive area) of CyclinA, p21WAF1, C-myc proteins and CyclinA mRNA,p21WAF1 mRNA were detected respectively by computerized image-analysis in normal skin epidermis, pathological skin scar epithelium and scar carcinoma tissues. All data was input into the computer and statistically analyzed with SPSS (16.0) software.Results:(1)C yclinA and its mRNA showed strong positive expression in scar carcinoma tissues, negative or weakly positive expression in normal skin epidermis and pathological skin scar epithelium. Compared with normal skin group and skin scar group, the expression (optical density and positive area) of CyclinA and its mRNA in scar carcinoma group were statistically significant (P<0.01). No statistic differences of their expression were measured between normal skin epidermis and pathological skin scar epithelium.(2) p21WAF1 protein and its mRNA showed strong positive expression in scar carcinoma tissues, negative or weakly positive expression in normal skin epidermis and pathological skin scar epithelium. Compared with normal skin group and skin scar group, the expression (optical density and positive area) of CyclinA and its mRNA in scar carcinoma group were statistically significant (P<0.01). No statistic differences of their expression were measured between normal skin epidermis and pathological skin scar epithelium.(3)C-myc protein showed strong positive, positive and weakly positive expression in scar carcinoma tissues, pathological skin scar epithelium and normal skin epidermis respectively. Each of two groups was statistically different (P<0.05). (4) Correlated analysis showed that there was positive correlation between the expression levels of CyclinA and CyclinA mRNA, p21WAF1 and p21WAF1 mRNA, CyclinA and p21WAF1 proteins, CyclinA and C-myc proteins, CyclinA mRNA and p21WAF1 mRNA in skin scar carcinoma tissues. Conclusions:(1)The high expression of CyclinA,CyclinA mRNA and C-myc may be related to the occurrence of skin scar carcinoma. (2) Skin scar carcinoma may be diagnosed in the earlier period by detecting the expression of C-myc protein. (3)There was the abnormal expression of Cyclin A and p21WAF1 not only on protein levels, there was abnormal expression but also on gene levels in skin scar carcinoma tissues.
