| Objective:To investigate the discipline and mechanism of 42℃hyperthermia on drug sensitivity of DDP by studying on the effect of 42℃hyperthermia on human ovarian cancer SKOV-3 cells in Vitro.Method:1. Cultured human ovarian cancer cells (SKOV-3) in Vitro. Detected the effect of DDP on human ovarian cancer cells proliferation by tetrazolium-based colorimetric assay.2. Through tetrazolium-based colorimetric assay measured growth inhibition rate of human ovarian cancer cells under different conditions, including 42℃hyperthermia, simple chemotherapy and hyperthermia chemotherapy combined with different sequential modes.3.Analyzed interaction index using jin's formula and estimated the inhibition effect of 42℃hyperthermia combined with chemotherapy on human ovarian cancer cells.4. Morphological changes were observed by inverted microscope.5. Measured the change of cells cycles using flow cytometry.Results:1.42℃hyperthermia and single drug also have inhibition and killing effect on human ovarian cancer cells (SKOV-3)individually.2. When combined 42℃hyperthermia with drug chemotherapy with different sequential modes, inhibition is stronger than using hyperthermia or chemotherapy individually(P<0.001). Especially, the inhibition effect is greatest when using hyperthermia and chemotherapy at the same time(P<0.001).3.42℃hyperthermia reduces the proportion of S phase cells by analysis cells cycles.4. Simple DDP reduces the human ovarian cancer cells at S phase.5. Compared with chemotherapy alone, the proportion of S phase cells reduces and the proportion of G1 phase cells increases when using chemotherapy prior to hyperthermia, hyperthermia prior to chemotherapy or chemotherapy and hyperthermia at the same time. Especially, the range of decrease and increase is maximal when using chemotherapy and hyperthermia at the same time.6. Cisplatin alone the proportion of the S cells decreased from 77.25% to55.10%.7. With the chemotherapy alone group and compared hyperthermia, The role of chemotherapy combined heat so the proportion of S-phase cells decreased from 77.25% to 5.03%,and G1 phase cells increased from thel.32% to 90.09%Conclusion:1.42℃hyperthermia has an apparent inhibition effect on human ovarian cancer cells(SKOV-3) proliferation and is a novel method of clinical treatment ovarian cancer.2.42℃hyperthermia combining with DDP chemotherapy has stronger inhibition effect on human ovarian cancer cells(SKOV-3) proliferation. And when using hyperthermia and chemotherapy at the same time, the inhibition effect is most obvious.3.42℃hyperthermia,single drug and 42℃hyperthermia combining with DDP chemotherapy may block the SKOV-3 cells at S phase and induce the SKOV-3 cells apoptosis.4. When 42℃hyperthermia combining with DDP chemotherapy, the DDP has stronger inhibition effect on human ovarian cancer cells(SKOV-3) proliferation. And when using hyperthermia and chemotherapy at the same time, the inhibition effect is most obvious. The mechanism may have a relationship with cells cycles interference. |
