The Effect Of Yin Nourishing And Antitorlc Capsule On The Expression Of Heat Shock Protein 70 And GR In Kidney Tissues In Rats With Radiation Injury By X-ray

Objective:To explore the protection mechanism of Yin Nourishing and AntitoxicCapsule's to X-ray injury in rats, by observing the effect that Yin Nourishing andAntitoxic Capsule have on expression of heat shock protein 70(HSP70) andglucocorticoid receptor(GR) in kidney tissues in rats with radiation injury. It canfurthermore provide the evidence for clinical application of Yin Nourishing andAntitoxic Capsule(YC).Methods:Experiment 1: Effect of Nourishing Yin Antitoxic Capsule on theheomgram in rats with radiation injuryFourty SD rats set up four groups at randoml: the normal group(A),theradiation group(B),the radiation and Nourishing Yin and Antitoxic group(C),the radiation and GuiFuDiHuang group(D),each group include ten rats. Then, it need to pulverize YC again for filtering 100-meshwork, and make it for 200mg/mL suspl with sonic oscillation by using distilled water. GuiFuDiHuang pillshave made to the same concentreation like Yc's too. Group A are treatmented2.5ml/day normal sodium to lavage before four days experiment. Group B alsohave been administrationed of 2.5ml/day normal sodium to lavage before fourdays X-ray radiation. Group C have been administrationed of2.5ml/day(condense for 0.2g/ml) Yc to lavage before four days X-rayradiation.And group D been administrationed of 2.5ml/day(condense for 0.2g/ml)GuiFuDiHuang pills to lavage before four days X-ray radiation. All groupsexcept group A have received same dose of X-ray radiation. Groups A,B,C,D got continuation of lavage for ten days.All animal are sacrificed after sixth dayby X-ray radiation,and the blood serum from heart are as samples to test.Experiment 2 : The Effect of Yin Nourishing and AntitoxicCapsule on the expression of Hot Shock Protein 70 and GR InKidney Tissues in RatsFourty SD rats set up four groups at randoml: the normal group(A),theradiation group(B),the radiation and Nourishing Yin and Antitoxic group(C),the radiation and GuiFuDiHuang group(D). All animal sacrificed after by Ycand GuiFuDiHuang pills administration 10 days. Observeing the expersion ofHS70 and GR in kidney in rat with radiation injury by X-ray, which provideeffect of YC. The experiment mode of duplicated like above experiment 1. Thenget kidney tissues, and determine the expression of HSP70 and GR usingwestern blot method.Results:1,Animal generally condition and the WBC,RBC and HGB of content in blood serum : Compared with the A group ,the contens of WBC,RBC and HGBin group B,C,D have a significance decrease(P<0.01). The clothing hair ofgroup C rats have a little loose and more than group B in food-intake andactivity amount, in contrast, which less than group A. The contens of WBC,RBC and HGB in group C Compared with the group B have a significancedifference(P<0.05). the clothing hair drop in group D less in group B, and thesedes is sofe shape in group D, which contens of WBC,RBC and HGBCompared with the group A,B have a significance difference (P<0.05),andhave no significance difference with group C. Illuminate the acute radiationinjury rat mode was success by intake 6 G y X-ray.2,The expression of HSP70 and GR in kidney : The hsp70 had apparentexpression in group B,group C and group D, with highest expression in group B,followed by group D, and lowest in group C. While GR had obviously higherexpression in group C, and group D ranked second.Conclusion: Yin Nourishing and Antitoxic Capsule can significantly reduce thehsp70 expression in kidneys of radiation injury by X-ray in rats, butGuiFuDiHuang pills have less function in doing so, which reflects the principleof TCM differentiation .Yin Nourishing and Antitoxic Capsule can improve GRexpression of radiation injury by X-ray in rats.
