Objective:To evaluate the efficacy of compound yolk emulsion against acute radiation oral mucositis through a randomized controlled trial for head and neck cancer patients treated by chemotherapy and radiotherapy. Methods:From March 2009 to January 2010,a total of 106 patients with newly diagnosed stageⅠ~Ⅳhead and neck cancer were randomly assigned to including 31 in the prevention group,42 in the treatment group and 33in the control group.All patients received current radiotherapy or chemo-radiotherapy. Radiation dose was more than 60Gy in the three groups.Intravenous infusion of compound yolk emulsion was started when radiation started one day in the prevention group and when oral mucositis occurred in the treatment group, which was given until the end of radiotherapy. Criteria of RTOG was used to evaluate acute oral mucositis and pain degree. Results:In the prevention group,the treatment group compare with the control group, the incidence for grade 3,4 mucositis and grade 2 developes to grade,which are the statistical significances(P<0.05).The prevention group had a longer average interval of oral mucositis when comparing with the treatment group and the control group, there are the statistical significances(P<0.05).For higher than grade 2 pain, the three groups were different from ach other. With the incidence rate of serious oral mucositis,smoking history is higher than no smoking history, chemo-radiotherapy is higher than radiotherapy,IMRT is lower than 3-DCRT, which are the statistical significances(P<0.05). Conclusions:Prophylactic or therapeutic use of compound yolk emulsion may postpone and decrease the severity of acute radiation oral mucositis, which can be profit to boost patients'quality of life during radiotherapy. Smoking history and chemotherapy are high risk factors of serious radioactivity oral mucositis.
