AIM:To investigate the expressions of CrkL mediated signaling proteins and their interventions by isoproterenol(ISO) in non-infarcted myocardium in rats. METHODS: MI and sham-operation were established by Litwin's method. Seven days after operation, surviving rats were divided into MI group(n=10),Sham group(n=6), MI+ISO group ( n=15) and Sham+ISO group(n=9), which were treated with physiological buffered saline(PBS) and ISO respectively. The expression of p-CrkL and C3G protein in non-infarcted myocardium was detected by Western blot. RESULTS:Twelve weeks after treatment, IOD of p-CrkL was (0.88±0.19) in MI group , (0.64±0.26) in Sham group, (1.20±0.16) in MI+ISO and (0.72±0.21) in Sham+ISO group respectively ; IOD of C3G was (1.14±0.29) in MI group, (0.90±0.10) in Sham group, (1.51±0.18) in MI+ISO and (0.97±0.26) in Sham+ISO group respectively. The expression of p-CrkL and C3G protein in non-infarcted myocardium was significantly higher in MI group as compared with Sham group respectively(both P <0.05); In MI+ISO group as compared with Sham+ISO group respectively(both P <0.05); and in MI+ISO group as compared with MI group respectively(both P <0.05); However, there was no any difference in the expression of p-CrkL and C3G protein in non-infarcted myocardium between Sham+ISO group and Sham group respectively(both P>0.05). CONCLUSION: The expression of p-CrkL and C3G protein was significantly increased in non-infarcted myocardium after MI, when given ISO, the expression could be further significantly increased. However, the significances of CrkL-C3G pathway proteins in post-infarction cardiac remodeling still remain to define.
