Aim: To study the potential effect and mechanism of simvastatin on connective tissue growth factor expression in the liver of diabetic rats.Methods:Thirty rats,4-months-old,male SD,were randomly separated into two groups:group A(normal control group,10 rats) and group B(20 rats),Diabetes(group B) was induced by injection of streptozotocin. Then the group B were randomly selected as group B1 (diabetic control group,10 rats) and group B2 (diabetic simvastatin group,10 rats). Group B2 was administered orally simvastatin (5mg·(kg- 1·d- 1 )), as well as the other groups were administered the same dose saline for 14 weeks. Then body weight,fasting blood sugar,serum triglyceride,serum total cholesterol,serum alanine aminotransferase and aspartate aminotransferase will be investigated.liver lesions were evaluated by HE staining. Immunohistochemistry for CTGF was performed by SABC technique.Results: To contrast with group A,the decline in body weight and rise in plasma glucose,total cholesterol and triglyceride of group B1 were significant(p<0.05),and group B2 had the same changes in body weight and plasma glucose,but no the same difference between B1 and B2.Group B2 had the lower level of total cholesterol and triglyceride than group B1(p<0.05). The difference of alanine aminotransferase and aspartate aminotransferase among three groups were not evident. Light microscopy in HE staining showed the hepatis lobules structure of three groups were limpid,and liver cell cord were in order, while a few lipid vacuoles can be seen in some hepatic cell . the CTGF expression in liver of group B1 and group B2 was increased significantly compared with the group A(p<0.05), but the increasing of group B1 was more significant than group B2 (p<0.05).Conclusion: The CTGF expression was increased significantly in the early stages of liver injury in diabetic rats, and simvastatin can suppress this change .oxidative stress and antioxidation may be the major role in this procedure.
