High recurrence rate after operation is its striking feature. Apigenin has many pharmacological activities,such as decreasing blood pressure,sedative,anti-bacterial and estrogen-like effects and so forth.Recent years, it makes the oncological scholars notice because of its inhibitory effect on many tumors. The study was to investigate the effect of apigenin on bladder cancer and its mechanism by the trials in vitro to the bladder cancer cell line BIU-87, and find a new way of preventing bladder cancer from relapse for the patients of reserving bladder in postoperative and treating advanced bladder cancer.PART ONE APIGENIN'S INFLUENCE ON THE BLADDER CANCER CELL LINE BIU-87 IN VITROObjective: To investigate the apigenin's influence on the proliferation of the bladder cancer cell BIU-87 in vitro.Methods: The cells'viability was detected by MTT assay in different time and different concentrations.Results: MTT showed that apigenin's inhibitory effect was ehanced with increasing time and concentration. There were significant differences among different concentrations in the same time and among different time in the same concentration(P﹤0.05).Conclusion: Apigenin can inhibit the proliferation of bladder cancer BIU -87 cells effectively in a dose- and time-dependent manner.PART TWO APIGENIN-INDUCED APOPTOSIS OF BLADDER CANCER CELL BIU-87 AND ITS MECHANISMObjective: To investigate the effect and the possible mechanism of apigenin on bladder cancer cell BIU-87 in vitro.Methods:①the cell cycle was measured by flow cytometry;②the cell apoptosis was observed by flow cytometry with Annexin V-FITC/PI double staining;③the expression of Survivin mRNA was detected by RT-PCR;④and the expressions of Survivin Protein and Caspase-3 Protein were detected by immunocytochemistry SP assay.Results:①After 72 hours of 22.03μmol/L apigenin treatment,it could arrest the BIU-87 cell in G2/M phase which was measured by flow cytometry, and we could find a obvious apoptosis peak.②The cell apoptosis rate was increeased significantly which was showed by flow cytometry with Annexin V-FITC and PI(P﹤0.05).③Compared with the control group, the expression of Survivin mRNA was significantly decreased(P﹤0.05), and so was the expression of Survivin protein(P﹤0.05), while that of Caspase-3 protein was markly upregulated(P﹤0.01) in the apigenin group.Conclusion: Apigenin can induce the apoptosis of BIU-87 cells.
