| Objective : To investigate the effect of a traditional Chinese compound,Jiejufushening(Radix astragli;Radix rehmanniae;Rhizoma coptidis;Radix et Rhizoma salviae multiorrhiae;Rhizoma Curcumae longae;Radix puerariae lobatae,etc)(JJFSN) on proliferation and secretion of extracellular matrix(ECM) of rat mesangial cells(MC).Methods : MC was cultured under high glucose condition in vitro .Sera containing different dose of JJFSN were prepared. The effects of the drug-containing sera on MC cell proliteration activity were assessed by the methylthiazoletetrazolium(MTT) assay. Cell cycle was detected by flow cytometry(FCM). And the concentration of collagen typeâ…£(Colâ…£) and laminin(Ln) in the supernantants were detected by radioimmunoassay (RIA).Results : JJFSN pharmacological serum significantly(P<0.05) inhibited MC proliferation activity under high glucose condition, in a dose and time-dependent manner.And after 48h treated in the high dose JJFSN pharmacological serum group,the OD value was even significantly lower than that in the normal glucose group(NG group) at the same time(P<0.05). FCM shows that JJFSN pharmacological serum increased the cell population in G0/G1 and reduced that in S-phase. Furthermore, high dose JJFSN pharmacological serum increased MC apoptosis. Meanwhile, compared with the high glucose group (HG group), the concentration of Colâ…£and Ln declined in different concentration JJFSN groups(P<0.05), in a dose-dependent manner. There was no significant difference among JJFSN high dose group and the NG group.Conclusions:JJFSN can reverse the influence of high glucose on MC proliferation activity, modulate cell cycle progress and induce cell apoptosis. And, it also can inhibit the secretion of Colâ…£and Ln. It implies that JJFSN might be a potential therapeutic agent for the prevention of DN in the early stage. |
