| Objective: By observation the effect of Edaravone Injection on the expression of MMP-9 in the brains of rats with Urokinase, reach the therapeutic mechanism of Edaravone towards hemorrhage prognosis after Infarction Treated With Urokinase..We examined the mechanisms by which Edaravone may help to suppress Urokinase-related hemorrhage.Method: Healthy male Sprague Dawley (SD) rats weighing 300 ~ 350g, making in rat middle cerebral artery occlusion model by autologous blood clot. after the success of model-making, rats were randomly divided into three groups, each group has 20 rats .Group UK was intravenously injected with 40000U/kg UK . Group CI received no treatment. Group UK+ED was intravenously injected with 40000U/kg UK plus 3 mg/kg Edaravone. Plus the 20 rats as sham-operated group. After 24 hours of modeling all the surviving rats were killed .We assessed the hemorrhage volume by pathological and spectrophotometer. The activity of MMP-9 in the brain was assessed by immunohistochemistry. We also studied the activity of mRNA of MMP-9, by PT-PCR.Results: (1) The results of brain histopathology in each group: Group UK has 7 of cerebral hemorrhage and group UK+ED has 4 of cerebral hemorrhage. Group CI has 1 of red blood cell leakage. There were no obvious pathological changes in sham-operated group (2) The results of MMP-9 and MMP-9 mRNA expression in each group of rat brain tissue: MMP-9 is highly expressed in brain tissue in group UK, contrast with the group CI there was significant difference (p <0.05).MMP-9 in group UK+ED expressed relatively less, there was significant difference (p <0.05). Like the expression of MMP-9, the expression of MMP-9 mRNA is similar in each group.Conclusion: Edaravone significantly reduced the expression of MMP-9 mRNA and MMP-9 in the brains of rats with Urokinase .Edaravone may reduce hemorrhagic transformation. |
