Objective: To study the distribution of the fibrinogen FGB-448G/A gene mutation in the CI group,to the mutation's impact on plasma fibrinogen and the genetic correlation between FGB-448G/A gene polymorphism and cerebral infarction.To analyze the relation between high plasma fibrinogen and the traditional risk factors of CI and find the new possible predictors for the primary prevention of CI.Data and methods: collect the informations of the 81 patients with CI(53 man and 28 women, their average age is 57.99±10.56 years old)and the control group of 76 patients(42 man and 34 women, their average age is 54.70±11.37years old). Analysed by PCR-RELP (polymerase chaine reaction of restricted fragment length polymorphism) ,FGB-448G/A gene polymorphism have three kinds of genotypes in fibrinogen FGB-448G/A gene polymorphism: homozygous(A/A),homozygous mutation(G/A) and wild type(G/G).The concentration of fibrinogen,glucose,lipids and hepatic,renal function were measured by conventional.Results:1. The mean fast plasma Fg concentration in CI group were 4.4351士0.99559g/L, higher than that of the control group ,3.8667士1.05173g/L, The difference is significant (P=0.001).2. The frequencies of the genotypes were: A/A0.02,G/A0.38,G/G0.60 in CI group and0.04,0.42,0.55 in the control group respectively,there were no significant difference (P=0.492).The frequencies of A allele in CI group were 0.237 and 0.204 in the control group, there were no significant difference (P=0.573) too.3. The mean fast plasma Fg concentration in G/G gene type and G/A,A/A gene type were 4.1257±1.19370g/L,4.2071±1.26693g/L respectively, there were no significant difference (P=0.636).4. According to Spearman correlative analysis and logistic Logistic-regression analysis and Stepwise method,the fast plasma Fg concentration was affected by age,the history of stroke and DM(the regression coefficient respectively was 0.042,2.216,-2.291 ).Conclusion: The FGB-448G/A polymorphism of fibrinogen may not be associated with plasma fibrinogen levels.The plasma fibrinogen level were related with age and the history of stroke and DM. |
