Objeetive: Colorectal cancer including the colon cancer and rectal cancer is one of the most common malignant tumors of digestive tract. In recent years, with the development of economic, the improvement of people's living standards and the change of lifestyles and living conditions, the incidence and mortality rates of colorectal cancer are increasing. The purpose of this study was to detect the expression level of Twist in colorectal cancer and normal tissue. At the same, ARF, E-cadherin are detected together, further to explore the relationship of Twist and ARF, E-cadherin protein with clinic-pathological indicators, and its function and mechanism at the diagnosis and prognosis of malignant tumors.Methods: 60 cases of colorectal carcinoma and 60 cases of normal colorectal tissues were collected in the Fourth Hospital of Hebei Medical University from June 2009 to December 2009 who took operations there. Colorectal cancer and normal colorectal tissues were collected in each of patient. Specimens were placed in 10% neutral formalin fixed and buried in paraffin, 4um serial section standby. All specimens were examined respectively Twist, ARF E-cadherin protein by immunohistochemical method. and the experiment result were dealed by spss15.0 statistical data analysis package.Results:1 The expression consequence of TwistExpression of Twist in 60 carcinoma and normal colorectal tissues were detected by IHC, the positive expression rates were 76.7%(46/60), 30.0% (18/60) respectively. The expression of Twist in carcinoma group was higher than nomal group (P<0.05) .Expression of Twist wasn't correlated with age, sex, tumor position (P>0.05), but correlated with pathological grade, the depth of the tumor invaded the bowel wall, condition of the lymph node metastases and Dukes stage (P<0.05).2 The expression consequence of ARFExpression of ARF in 60 carcinoma and normal colorectal tissues were detected by IHC, the positive expression rates were 30.0%(24/60), 73.3% (44/60) respectively. The expression of ARF in carcinoma group was lower than nomal group (P<0.05) .Expression of ARF wasn't correlated with age, sex, tumor position (P>0.05), but correlated with pathological grade, the depth of the tumor invaded the bowel wall, condition of the lymph node metastases, Dukes stage and distant place metastasis (P<0.05).3 The expression consequence of E-cadherinExpression of E-cadherin in 60 carcinoma and normal colorectal tissues were detected by IHC, the positive expression rates were 13.4%(8/60), 60.0% (36/60) respectively. The expression of E-cadherin in carcinoma group was lower than nomal group (P<0.05) .Expression of E-cadherin wasn't correlated with age, sex, tumor position (P>0.05), but correlated with pathological grade, the depth of the tumor invaded the bowel wall, condition of the lymph node metastases and Dukes stage (P<0.05).4 The relationship of Twist, ARF and E-cadherinThere were 10 cases in the colorectal carcinoma group where the expression of Twist and ARF was positive simultaneously, the simultaneously negative were 8 cases. Spearman correlation showed that Twist had a remarkably correlation with ARF in colorectal carcinoma (r=0.806, P<0.05). There were 3 cases in the colorectal carcinoma group where the expression of Twist and E-cadherin was positive simultaneously, the simultaneously negative were 7 cases. Spearman correlation showed that Twist had a remarkably correlation with E-cadherin in colorectal carcinoma (r=0.754, P<0.05).Conelusion:Twist may be the biological parameter of colorectal carcinoma ,as well as determine the course of disease in patients with colorectal disease- specific indicator of prognosis.P53 protein , E- cadherin plays an important role in the proeess of colorectal cancer growth,invasion and metastasis,Twist probably inhibits apoptosis and promotes tumor cell formation by regulating pathway of ARF/MDM2/P53, meanwhile impacts expression of E-eadherin to promote situation of EMT, and participates in the oecurrence and metastasis of colorectal cancer.
