Viral pneumonia is a commonly type and about 50% in clinics.Respiratory syncytial pneumonia(RSVP) is the most common type among children's viral pneumonia, it threatens seriously the life and health of children. The modern medicine does not have special treatments for the RSV pneumonia. And currently, an RSV vaccine is not yet available. However, the Chinese medicine treatment has certain superiority.IFN-α/βplay an important role in the respiratory virus infections, it as the important component of natural immunity can inhibit virus replication in the early stage of the virus infection. However, RSV infection rarely induced interferon, and in many times, it could reduce anti-viral effect of IFN.Clinical and experimental researches show that JOL(Jinxin Oral Liquid) have anti-RSV infection effect, However, the precise mechanism of its anti-RSV properties is unknown. It is extrapolated that JOL oppresses RSV by up-regulating the the expression of IFN-1 of parasitifer. This research is based on the previous study.Objectives:1.To research a model building of rat RSV pneumonia.2.To research the regulating action of JOL Liquid on IFN-1 expression of rats infected by respiratory syncytial virus.Methods:With the methods of cell culture, laboratory animal science, molecular immunology, ELISA assay, we detected the alteration of IFN-1 expression in rats. Results were analyzed with statistics software SPSS 13.Results:Clinieal manifestations, for example:increased body temperature, decreased activity, breathlessness, mussyhair and occasional cough gradually emerged in RSV-infeeted rats. There were no significant differences in the temperature change between RSV-infeeted group and control group. RSV-infeeted group have little effect on general state of health. But there were significant differences in average body weight. Lung biopsy from RSV-infeeted group showed typical inflanunation under light microscope. And infected lung tissue could separate RSV virus. It is suggested that RSV-infected models were successful.The IFN-αexpression of prevention group and JOLM(Jinxin Oral Liquid Middle dose) group was higher than model group (P<0.05); The IFN-βexpression of JOLH(Jinxin Oral Liquid High dose) group and JOLL(Jinxin Oral Liquid Low dose) group was higher than model group (P<0.05); The IFN-βexpression of prevention group and JOLM group and ribavirin group was higher than control group (P<0.05),which was also higher than JOLL group (P <0.05).Conclusions:1.With the methods of driping RSV liquid into the nose of young rats, we can build a model of rat RSV pneumonia.2. JOL could play the antagonistic role of the RSV.3. JOL can upregulate the expression of IFN-â… of parasitifer with RSV infection, which make IFN-â… plays a role of antivirus. This is also one of the anti-RSV mechanisms of JOL. |
