Study the effect of estradiol and 5-HT by acupuncture some acupoints of neutered big rats;And research the changing of sex hormone,index of Kupperman and grade of HAMD after acupuncture some acupoints to treat depression in peri-menopausa period.Experiment of animanl:The 32 rats according to body weight were randomly divided into four groups,they are normal control group(A), Model group (B);,acupuncture treatment group (C) and estrogen treatment group (D). In addition to A group, the other three groups rats were ovariectomized surgery. Administration after a week. A, B groups were given normal saline gavage; C group was given acupuncture every other day; D group received estradiol intramuscular injection every other day. Acupuncture and administration 30 times. after 24 h treatments,then taking blood ang separating the hypothalamus.Detect serum estradiol levels by Chemiluminescent and test 5-HT content in Hypothalamic by ELISA.Clinical trial:The 30 patients with gynecological malignancies, or after radiotherapy and chemotherapy in patients with perimenopausal depression were randomly divided into treatment group and control group. Detect the levels of serum E2, FSH, LH with Chemiluminescence and test the levels of serum INHB by ELISA.The levels of serum E2 and hypothalamic 5-HT of model group decreased, accord with, laboratory indices changes of perimenopausal depression. Acupuncture and estrogen have the role to rise serum E2. But the role of acupuncture is lower than estrogen. There is positive correlation between serum E2 and hypothalamic 5-HT.Compare the serum E2 of treatment group and control group before treatment,we find that there is no difference.After three monthes,compare all the groups,we find that the serum E2 of treatment group is the highest, but control group's is the lowest, and they have difference. Compare treatment groupafter and before treatment,we find that there is difference in serum E2. It showes that acupuncture can regulate the serum E2.Compare the serum FSH and LH of treatment group and control group before treatment,we find that there is no difference.After three monthes,compare all the groups,we find that the serum FSH and LH of treatment group are the highest, but control group's are the lowest, and they have no difference.Compare the symptom,for example:hot perspire and sweating, sleeplessness, depression, of treatment group and control group before treatment,we find that there is no difference.After three monthes,compare all the groups,we find that, and they have difference. According to Kupperman index, HAMD factor score to assess the treatment effect.the effective.rate is 87%,80% respectively. It show that acupuncture can improve the symptom of perimenopausal depression.Acupuncture points can smooth the liver and reinforce rssence of the kidney, conditioning Chong and Ren, regulating peri-menopausal period of serum E2 levels in patients with depression,and regulate Hypothalamic-pituitary-ovarian axis. Reconstructing part of the estrogen negative feedback role in influencing the perimenopausal depression of the nervous system functional state.Thereby reducing the incidence of depression of the peri-menopausal period.
