Objective TO investigate Effects of retrograde coronary sinus perfusion vascular endothelial growth factor (VEGF) on L-type current in rabbit myocardial infarction model, so as to explore the basis of VEGF treatment for antiarrhythmia.. Methods Forty-five rabbits were randomly divided into three groups, conventionalgeneral anesthesia and open the thorax. Groupâ…¡(nature saline control group) clamping the main stem of left ventricular coronary. nature saline were retrograde perfused through coronary sinus. Groupâ…¢(VEGF treatment group), VEGF were used.The instances of malignant arrhythmia were recorded. Single ventricular myocyte were isolated from the epicardial zone of the infarcted region, whole cell patch clamp technique was used to record L type calcium(Ica-l.) simultaneously, the level of VEGF, nitric oxide, (NO) and cardio troponin T, (TnT) before and 120min post operation were tested. Results:Preoperative there were no significantly difference in VEGF, NO and TnT concentration among three groups (p>0.05).120-min postoperation, in groupâ…¡andâ…¢, TnT was significantly elevated and higher than that in groupâ… (p<0.05 or 0.01),but groupâ…¡was lower than groupâ…¡(p<0.05). The instance of malignant arrhythmia and peak of the current density(Mv) in groupâ…¡were higher than that inâ… andâ…¢(p<0.05 or 0.01).The level of VEGF and NO were significantly higher than that in groupâ… andâ…¡(p<0.05 or0.01)Conclusions Retrograde coronary sins perfusion VEGF could increase the NO level,regulate the Ica-L current and thus decrease the incidence of maglignant arrhythmia. |
