Objective To investigate whether alpha-1 antitrypsin is present in normal human aorta, ascending aortic aneurysm and stanford A type aortic dissection(TAAD) tissues and whether there are some expression differences of alpha-1 antitrypsin levels among them, and pave a way for advanced research of its potential protective effect for aortic wall structure.Methods Histology, Western blotting and semi-quantified RT-PCR were included to test whether alpha-1 antitrypsin is present in normal human aorta, ascending aortic aneurysm and TAAD tissues.16 cases TAAD tissues,10 ascending aortic aneurysm tissues and 8 normal ascending aorta tissues were included in the study.Results Alpha-1 antitrypsin expression was strongly increased in TAAD and ascending aortic aneurysm tissues compared with normal aorta(0.285±0.010,0.153±0.011,0.102±0.010, p<0.05). Alpha-1 antitrypsin increased more significant in ascending aortic aneurys than in TAAD (0.285±0.010,0.153±0.011, p<0.05). The positive results of RT-PCR tested that there were endogenous alpha-1 antitrypsin produced in aortic wall and mRNA expression levels of them were highest in ascending aortic aneurym and lowest in TAAD (2.192±0.133, 1.213±0.156,0.672±0.101, p<0.05). Alpha-1 antitrypsin was present throughout the entire lamina of ascending aorta. Conclusion Alpha-1 antitrypsin is present in normal human aorta, ascending aortic aneurysm and TAAD tissues and there are some expression differences among them. Alpha-1 antitrypsin presents a potential pathway for protection of aortic wall structure.Objective To investigate the blood vessels structural characteristics of normal human ascending aorta, ascending aortic aneurysm and Stanford A type aortic dissection(TAAD) and discuss the protective effects of alpha-1 AT for Stanford A type aortic dissection.Methods HE staining method, Vigesion staining method and TdT-mediated dUTP nick end labeling (TUNEL) method were included to test the blood vessels structural characteristics of normal human aorta, ascending aortic aneurysm and TAAD.16 TAAD tissues,10 ascending aortic aneurysm tissues and 8 normal ascending aorta tissues were included in the study.Results The vessel walls of normal ascending aortae and ascending aortic aneurysms have similar structure, consisting of intima, tunica media and adventitia. However, elastic fibers become more thinner in ascending aortic aneurysm tissues, even existing absence of elastic fibers and it is relatively normal in outer side, and the conspicious hyperplasy of smooth muscle also could be observed. TAAD ascending aortae show conspicuous collapse of elastic and collagen fibers,thinner, inflammatory cells infitration and blood cells. The results of TUNEL show that there are many apoptosis cells in TAAD tissues and not obvious apoptosis in normal ascending aortae and ascending aortic aneurysm tissues.Conclusion Compared with ascending aortic aneurysm and normal ascending aortae, TAAD ascending aortae show conspicuous collapse of elastic and collagen fibers, thinner, inflammatory cells infitration and apoptosis of smooth muscle cells.
