Preface:The incidence of stress urinary incontinence(SUI) reaches up to 10% in female group, and to 17.1% in postmenopausal female group. When the abonominal pressure increases, the urine can be outside of urethra out of control, which has a great impact on women's health and life quality. It's not permitted or there's no need to do the experiment or study on human's body directly, so the establishment of animal model and evaluation of it can be used to research the human's disease on the animal's body. This research makes the procession of establishing an animal model much closer to the course of pregnancy and delivery of the pregnant women by imitating the pathogenesis of the human's SUI which has already been recognized. Identifying the animal model by sneeze test and urodynamic indicators, then detect pelvic floor muscle strength and determination of serum biochemical(LDH,CK), pelvic floor muscle acetylcholine receptor counts, RT-PCR was applied in different periods after the establishment of rat model to detect the expression of Caspase-3, and then do the histological identification (H&E staining). To research the relevant change in the model's pelvic fiber can provide a reliable, actual, steady and repeated anima model for the study of clinics.â… . PART ONEObjective:To establish rat model of stress urinary incontinence. Methods:To place a hyponome in the rat vagina for a long time to imitate the way in which birth trauma destructs the rat pelvic floor for building a SUI animal model and made sneeze test and urodynamics to evaluate the success of the rat model.Results:1,8 hours is the best time for building a SUI rat model.2,The successful incontinence was evaluated 72.5%(29/40) by the sneeze test. 3. By the urodynamics detection, the leakage of urine point pressure and the maximum bladder capacity in the model both decreased. Conclusion:The method by expanding the vagina can establish the SUI rat model successfully.â…¡. PART TWOObjective:To observe the change of injury in rat model's pelvic fiber.Methods:To research the SUI rat model by sneeze test and urodynamic indicators detection, then detect pelvic floor muscle strength and determination of serum biochemical (LDH, CK), pelvic floor muscle acetylcholine receptor counts, RT-PCR was applied in different periods after the establishment of rat model to detect the expression of caspase-3, and then do the histological identification (H & E staining).Results:Sneeze test showed positive rate and the largest bladder capacity, leak point pressure of the experimental group in SUI modeled rat decreased, pelvic floor muscle contraction strength declined, lactate dehydrogenase and creatine kinase showed a downward trend after previous rise, pelvic pubococcygeus acetylcholine receptors of group presented the recovery trend after some decline, the expression of caspase-3 of the pubococcygeus firstly increased and then decreased. HE staining results suggested that muscle fibers gradually increased after the first injury.Conclusion:Every index in the rat model after imitating dystocia showed exceptional and resumed to normal after 4 to 8 weeks.
