Objective:To observe the influence of different time of hypotension to myocardial emzyme and expression of cTnI in the rats under the threshold controlled hypotension. This research aims to make clear about the time limit for the continued safety of the threswhold controlled hypotension and the molecular mechanism of the occurrence and development of myocardial injury。Methods:24 rats were randomly divided into 4groups, each group with 6 rats:Group A(normal);Group C(sustained controlled hypotension), Group C set three different points sustained controlled hypotension:2h, 4h,6h.So 3 groups:C1,C2,C3.Use sodium nitoprusside (SNP) and esmolol (ESM) induce controlled hypotension. During the time of lowering blood pressure, recorded rats'vital signs and did blood gas analysis.24hours later, observed rats'neurobehavioral functions and survival rate after hypotension, recorded the blood gas analysis.Having a laboratory test of myocardial emzyme and detecting the cTnI with immunohisto chemical staining after 24 hours.The survival rate of the rats are also recorded.Result:1.The leve of LDH,CK,CK-MB in the group C1 is more higher than that in the group C2,groupC3 and gropu A (P<0.05),and LDH than 1000u/l;the level of LDH,CK,CK-MB in group C 2 is the highest among that in the group Cl,group A (P<0.05),and LDH, CK, CK-MB are more than 1000u/1;the level of LDH,CK,CK-MB in group C3 is the highest among that in the group C1,groupC2 and group A(P<0.05).2.The expression of cTnI in group C1 is higher than that in group C2,group C3 and group A(P<0.05);the expression of cTnI in group C2 is higher than that in group C1,group C3 and group A(P<0.05).The expression of cTnI in group C3 and is higher than that in group C1,group C3 and group A(P<0.05).Conclusions:Threshold controlled hypotension 55mmHg for 2 hours,the rat myocardial emzyme is increased significantly,Troponin I expression appears,The myocardial injury。With the further extension of time hypotension,Myocardial injury is worsened。...
