| Objective:To explore the regular expression of Matrix Metalloproteinase-9 (MMP-9),SOD,MDA and the neuroprotective effect of lycopene after focal cerebral ischemical reperfusion jury in rat.Methods:Totally 70 wistar rats were randomly divided into 3 groups,the sham-operated group,ischemica/reperfusion group (I/Rgroup 6h,24h,48h,72h,7d group after CIRP 2h) and the lycopene-treatment group(LP group 24h group after CIRP 2h).Rats were intraperitoneal injected with lycopene butter resin capsule resveratrol in lycopene-treatment group a week, the sham group and I/Rgroup was pretreated with normal saline for 7 days before the model of middle. The neurological defieit score was graded by Zea Longa's methods, the expression of MMP-9 by immunohistochemistry;the expression of SOD,MDA by colorimetry and the water content was calculated by dry and wet weight method.Results:1)According to the neurologieal defieit seores,the significant difference existed on the scores between the control group and the lycopene groups at 24h.2)The MMP-9 began to express on MMP-9 positive cells at 6h reperfusion in I/R group after CIRP, then it increased at 24h and reached peak level at 48h, after this,the level of MMP-9 began to decrease up to 7d.3) The water contens were significant different existed on the LP group and control group at 24h.4) The effect of SOD and MDA: SOD activity and MDA content in brain were determined respectively.The activity of SOD decreased immediately after operation,come to the lowest point after 24-48h,then increased in the following time, whereas the leaves of MDA came to the highest point after 48h,then decreased.There was significance difference between sham group and I/Rgroup(P<0.01).Conclusion:The expressions of MMP-9 incereased in the early stage of reperfusion and it decreased following stage.The activity of SOD decreased after operation, then increased in the following time, whereas the leaves of MDA came to the highest,then decreased.They have negative correlation.The lycopene can reduce the expression of MMP-9 after focal cerebral ischemia-reperfusion injury,can lighten the level of vasogenic brain edema,and improve the neurological defieit scores,so it have a neuroprotective effect on impaired neuron in cerebral isehemia-reperfusion injury. |
