Porcine serum protein is hydrolyzed by trypsin protease, neutral protease and alkaline protease to prepare hydrolyzed serum. Optimal hydrolysis conditions as follows:(1)The best conditions by trypsin protease for protein hydrolysis are temperature=54℃, time=2.4h, pH=8, the enzyme= amount=4480u/g substrate.The degree of hydrolysis is 14.83%. (2), The best conditions by neutral protease for protein hydrolysis are temperature=54℃, time=4.5h, pH=8, the enzyme amount=3489u/g substrate. The degree of hydrolysis is 16.90%. (3) The best conditions by alkaline protease for protein hydrolysis are temperature=50℃, time=5.3h, pH=9, the enzyme concentration= 1190u/g substrate. The degree of hydrolysis is 21.26%. Then, we found the order for dual-enzyme digestion enzyme that the trypsin protease after alkaline protease, the degree of hydrolysis is 25.23%.The result showed that the porcine serum protein polypeptide was mainly distributed in the following lOkDa by SDS-polyacrylamide gel electrophoresis. The product was separated into≤3KDa,3KDa-10KDa and≥10KDa parts by ultrafiltration device. The three parts of its component polypeptide share a percentage of total output of 38.1%,59.32% and 2.58%.Based on "Inspection and Evaluation of Technical Specification of Health Food (2003)" and compared of soybean polypeptide, the research register body weight of mice, serum urea nitrogen, liver glycogen, muscle glycogen and blood lactic acid. A number of indicators were measured to anti-fatigue effect in mice of the pig serum peptide. The results showed that swim test was positive, serum urea nitrogen, liver glycogen, muscle glycogen, blood lactic acid are all positive. The results show that porcine serum protein polypeptide is good at anti-fatigue effect.Finally, the research developed a health oral liquid by porcine serum protein polypeptide by thick, sweet, sour taste and jujube, medlar aroma. The formula is as follows:pig serum polypeptide 24%, L-carnitine 0.06%, jujube extract 25%, wolfberry extract 25%, sugar 8%, citric acid 0.1%, sodium citrate0.01%, xanthan 0.06%, ethyl maltol 3mg/kg, and sodium hexametaphosphate 0.8g/kg. The inspection results are as followed:moisture content 68%, protein content is 23.56%, ash content is 2.74, and soluble solids content is 31.32%. According to national standard inspection, we can get the microbial indicator of product as follows:no colonies, the total number of E. and pathogen detection is check out.
