Postmenopausal osteoporosis is a common primary osteoporosis, high incidence rate, has serious health threat to the medium age and older women. However, the pathological mechanisms have not remain clear, the effectiveness of drug prevention is not ideal.Traditional Chinese Medicine believes that "Kidney controls bone" and " Kidney weak, bones weak" and has a unique and rich theory and experience on the pathogenesis and treatment of bone disease, and accumulated a tremendous amount of Chinese medicine literature. Based on "Kidney controls bone" theory of Chinese medicine, osteoporosis and kidney are more relevant. Therefore, Bushenzhuanggu treatment of osteoporosis is the ultimate solution. In this study we use a Traditional Chinese Medicine treatment of senile kidney deficiency and joint alfacalcidol to treat osteoporosis of ovariectomized rats, also try to explore the mechanism of it.ObjectiveTo study the combined effects of Bushen Chinese Drugs and Alfacalciferol on bone mass index and metabolism biochemical markers of OVX osteoporosis rats.MethodsThe SD female rats osteoporosis model was established by the method of ovariectomy (OVX). The rats were divided into five groups:control (SHAM), ovariectomized (OVX), OVX administrated with Bushen Chinese Drugs (OVX +TCM), OVX administrated with Bushen Chinese Drugs and alfacalciferol (OVX+TCM+VD), and OVX administrated with nilestriol (OVX+CEE3). The drugs were administrated after 35 days of ovariectomy. All rats were sacrificed after 13 weeks of treatment. The serum E2, bone mineral density (BMD) of vertebrae and left femur were detected and morphological quantitative analysis of bone tissue of femurs was performed and was detected. The ratio in weight of uterus to body was calculated, and uterine slice was gotten for histological observation.Results1. OVX rat uterus wet weight, uterine weight/body weight, endometrial thickness and serum E2 level were significantly lower than those of SHAM, indicating OVX rats with both ovaries removed completely. Lumbar spine bone mass and bone histomorphometric, biomechanical properties of indicators of osteoporosis, were significantly lower than the SHAM group (P<0.01), serum ALP and urinary Pyd/Cr increased, the formation of high conversion pathological features and estrogen deficiency rat model of osteoporosis is succeeded.2. OVX+CEE3 groups show Lumbar spine bone mass and bone histomorphometric, biomechanical properties of bone and other indicates a marked increase compared with OVX group. Bone turnover markers are inhibited.3. OVX+TCM+VD group significantly increased bone mass, bone structure significantly improved performance due to improved fracture resistance of bone, bone formation and resorption biochemical markers reduced. OVX +TCM+VD group has superior prevention and treatment of osteoporosis than OVX+TCM group.4. Serum 1,25 (OH) 2D levels of OVX rats were significantly higher (P<0.01), and the emergence of lower serum calcium, low calcium and high levels of 1,25 (OH) 2D to become active bone resorption, One of the mechanisms pathological causes of osteoporosis. Nilestriol can restore blood calcium level,1,25 (OH) 2D levels drop, bone remodeling rate inhibited, as the effective mechanism of prevention and treatment of osteoporosis.5. OVX+TCM and OVX+TCM+VD group have no significant restoration of serum calcium, serum 1,25 (OH) 2D level remains high level. ConclusionsThe serum 1,25(OH) 2D in OVX rats significantly increased, possibly one of the pathological mechanisms leading to osteoporosis. Nylestriol has effection on estrogen deficiency-induced bone loss and bone structural changes. It could be related with the correction of low level of serum calcium and high level of serum 1,25 (OH) 2D. OVX+TCM+VD has the superior effection on osteoporosis in ovariectomized rats than the Chinese herbs alone. The mechanism of prevention and treatment of osteoporosis may be related to the role of multi-target of Bushen Chinese Drugs and high-level 1,25 (OH) 2D up 24,25 (OH) 2D, increased bone formation and bone mineralization.
