Developing A New Antithrombotic Drug And The Study Of Pharmacodynamics

Many traditional Chinese medicine (TCD) monomers have a specific and safe antithrombotic effect, but there is a higher dose needed in clinical cure because of its weak action. So it how to make these Chinese medicine monomers that effects on different ways and links of thrombosis composed reasonably, using associatly, increasing action, reducing side effect, is becoming more and more interesting on the medical field.Objects:①Selecting the combination of anti-thrombus traditional Chinese drug.②Observing the combination's effect on venous thrombosis.③Observing the combination's effect on microthrombosis.Methods:①Selecting the combination of anti-thrombus traditional Chinese drug on prothrombin time (PT),activated partial thromboplastin time (APTT),recalcification time (RT) and fibrinolysis in vitro.②Observing the combination's effect on venous thrombosis of ratand using orthogonal design to find the best dose of the combination.③Observing the combination's effect on microthrombosis of mouse.Results:①The anti-thrombus TCD combination ABC can lengthen PT,APTT,RT (P<0.01) and promote fibrinolysis obviously (P<0.01), and the effect is better that AB,AC and positive drug Ligustrazine (TMP) (P<0.05),but the effet on promoting fibrinolysis is similar as BC(P>0.05).②The anti-thrombus TCD combination (1/3×A+3×B+1/3×C) can inhibit the formation of venous thrombosis of rat obviously (P<0.01), and the effect is better that other combinations and TMP (P<0.05).③The dose (1/3×A+3×B+1/3×C) of anti-thrombus TCD combination can make the blood platelet count(BPC), PT, the concentration of fibrinogen(Fbg) and anti-thrombin-Ⅲ(AT-Ⅲ) of DIC mouse to recover(P<0.05), and at the same time, the anti-thrombus TCD combination also can make the damage of lung and kidney of DIC mouse to ease obviously or to recover.Conclusions:①Traditional Chinese drug (TCD) anti-thrombus combination that composed by A,B and C that have effects on inhibiting the activation of blood platelets, promoting fibrinolysis expand blood vessels, which has a good effect on anti-thromboxane and promoting fibrinolysis in vitro.②The TCD anti-thrombus combination can inhibit the formation of venous thrombosis of SD rat obviously, and the best dose is 1/3×A+3×B+1/3.③The TCD anti-thrombus combination has a good effect on microthrombus mouse that induced by LPS.
