Detect the effects of demethylation on the nuclear transcription factor expression profile of colon carcinoma cell line.(1) Applied demethylation reagent 5-Aza-Cdr of different concentrations to colon carcinoma cell HT-29, and detected the cell growth curve,cell cycle and apoptosis;(2) Based on the results above, we used high-throughout transcription factor assay to discuss the effects of 5-Aza-Cdr on the nuclear transcription factor expression profile of colon carcinoma cell line HT-29;(3) Analyzed the altered transcription factors, and verified a part of the results of transcription factor assay with western blotting.(1) 5-Aza-Cdr inhibited cell growth(P<0.01), induced G2/M phase arrest in cell cycle and promoted apoptosis (P<0.001) in HT-29 colon carcinoma cells;(2) 5-Aza-Cdr can induced changes of transcription factor expression in the nuclear of colon carcinoma cell HT-29.The up-regulated and down regulated transcription factors more than 1.5 times were amount to 25.(1) Demethylation reagent 5-Aza-Cdr role in colon cancer cell HT-29 can cause biological changes that were inhibition of cell growth, inducing cell cycle G2/M phase arrest, and promoting cell apoptosis.(2) Most of nuclear transcription factors of colon carcinoma cell HT-29 could not be regulate by epigenetics,a few of them could be regulate by epigenetics. The altered transcription factors belonged to functional categories of cell proliferation,cell differentiation, cell cycle controlling, cell apoptosis,growth and development, drug metabolism, hormone secretion, gene replication process and so on.(3) These altered transcription factors belonged to some important signal pathways. It provided a theoretical basis for analysis of regulation by epigenetic on signaling pathways in colon carcinoma cell.
