[OBJECTIVE]The pharmacokinetics difference between the traditional slice decoction and dispensing granule decoction was compared, taking the famous heat-clearing and detoxicating decoction, named Huanglianjiedu decoction as an experiment object. To discuss the feasibility of application of the latter in complex prescription and provide an evidence for its clinical practice.[METHODS]This research was divided into three parts as follow,1 The main index contents were determined in between two decoctions by HPLC, to ensure the contents of berberine and the baicalin are consistent before Intragastric administration to the rat.Huanglianjiedu decoctions used in the experiment are both self-made traditional slice decoction and dispensing granule decoction. The two decoctions are dealt with the sampling method, determine the contens of the berberine and the baicalin, then analyse the data by the SPSS12.0.2 The contents of berberine in the rat plasma was determined between two decoctions by HPLC. Compare the pharmacokinetic characters of berberine in the rat plasma after Intragastric administration of traditional slice decoction and dispensing granule decoction.3 The contents of baicalin in the rat plasma was determined between two decoctions by HPLC. Compare the pharmacokinetic characters of the baicalin in the rat plasma after Intragastric administration of traditional slice decoction and dispensing granule decoction. In the experument,36 healthy and male mice were selected, divede them into 6 groups at random,6 for each group, they are the low dose traditional slice decoction, the middle dose traditional slice decoction, the high dose traditional slice decoction, the low dose dispensing granule decoction, the middle dose dispensing granule decoction and the high dose dispensing granule decoction. Intragastric administration 7.5 g/kg,15 g/kg,30g/kg Huanglianjiedu decoctions to the six groups. Take the blood and determine the contents of berberine and baicalin in the rat plasma respectibvely. Analyze the data by the DASS2.1.1 and SPSS 12.0 softwares, then calculate the parameters and compare the C-Tcurve, the Tmax, Cmax, the bioavailability and so on between the two decoctions.[RESULTS]In the experiment of the pharmacokinetics of beiberine and baicalin, the berberine and the baicalin were detected in each group of the traditional slice decoction and the dispensing granule decoction. The C-T curve of the berberine and the baicalin are consistent between two decoctions, and the Tmax, Cmax and the bioavailability have no significant differences.[CONCLUSION]Administrated by the same dose of the traditional slice decoction and dispensing granule decoction to the mice, the berberine and the baicalin were detected in each group of both decoctions, and the C-T curves are consistent between the traditional slice decoction and dispensing granule decoction, and the Tmax, Cmax and the bioavailability have no significant differences. It provided a solid therial basis for the clinical use of dispensing granule decoction.
