| Objective:1. To confirm that transfection of nerve growth factor (Nerve growth factor, NGF) gene in rat bone marrow mesenchymal stem cells(rBMSCs )in diabetic rat bladder tissues can alive.2. To confirm that the NGF gene in rBMSCs in diabetic rats bladder tissues can stably express.Methods:1. To built the diabetic model of rats .2. BrdU-labelled immunohistochemistry was used to observe the rBMSCs of transfection with NGF gene.3. The BrdU-labelled rBMSCs of transfection of NGF gene was transplanted into the diabetic rats bladder tissues.4. BrdU-labelled immunohistochemistry was used to observe the growth of rBMSCs of transfection of NGF gene in the diabetic rats bladder tissues.5. We checked the infected rBMSCs about the expression of NGF mRNA and protein by RT-PCR and ELISA.Results:1. A single intraperitoneal infection of STZ successfully established rats model of diabetes.2. BrdU-labelled immunohistochemistry was successfully used to observe the rBMSCs of transfection of NGF gene.3. The BrdU-labelled rBMSCs of transfection of NGF gene was successfully transplanted into the diabetic rats bladder tissues.4. BrdU-labelled immunohistochemistry was successfully used to observe the growth of rBMSCs of transfection of NGF gene in the diabetic rats bladder tissues.5. We checked the expression of NGF gene by RT-PCR and ELISA.Conclusions:1. The infected rBMSCs can alive in diabetic rats bladder tissues.2. The NGF gene in rBMSCs in diabetic rats bladder tissues can stably express. |
