The paper has revised the pharmacopoeial standard of Oleum Viticis Negundo based on the Ch. P (2005). In order to determine short tandem repeats (STR) with a simple, rapid and low-cost method, the expert system for STR genotyping was studied based on spectroscopy combined with principal discriminant variate (PDV) algorithm.For the revision of pharmacopoeial standard of Oleum Viticis Negundo, we collected the Folium Viticis Negundo of different regions and harvest seasons, and then the Oleum Viticis Negundo were extracted by ourselves according to the established project. At the same time, we also purchased the Oleum Viticis Negundo from different factories with different Batches. The identification method of TLC, heavy metal limitation and GC assay method were added for the Oleum Viticis Negundo.The identification method of TLC was established based on the researches of different developing solvents, colour reagents and sample applications. The method is specific, steady, convenient and with good reproducibility according to the studies of methodology and determination of all the samples. It could be used for the identification of Oleum Viticis Negundo. According to the characteristic of Oleum Viticis Negundo and the heavy metal test of other essential oils, we chose the second heavy metal test in Ch.P appendix to determine the heavy metal in Oleum Viticis Negundo and then defined the limitation not more than 0.001%. Used GC to determine the samples of essential oil, and then combined with GC-MS, the index componentβ-caryophyllene for assay of Oleum Viticis Negundo was confirmed. The contents ofβ-caryophyllene in Oleum Viticis Negundo of different sources were determined and then the methodology and universality of the method were studied. The content range ofβ-caryophyllene was 25.44%-69.87%, and it should not less than 20.0% in officinal Oleum Viticis Negundo. The method is of good accuracy, reproducibility and durability. The analytic time is short and the adaptivity is wide. It can be used as pharmacopoeial method for determination ofβ-caryophyllene in Oleum Viticis Negundo.D16S539 locus, which has been widely applied in forensic medicine, was selected as the research object for the expert system of STR genotyping based on spectroscopy. Firstly, two genotypes with small difference in the total number of core repeat units were used to establish the PDV model based on near infrared spectroscopy. Then the expert system for the selected six genotypes was established based on the difference of their total number of core repeat units. The six genotypes were discriminated correctly and the accuracy of prediction samples was 100%.In order to make the STR genotyping simpler, ultraviolet spectroscopy (UVS)-PDV model was studied. The methodology was studied by using the genotypes having middle degree and the same total number of core repeat units. Firstly, the genotypes whose difference was 4 were selected to establish the model, and they could be discriminated correctly based on UVS. The genotypes with the same total number of core repeat units were also discriminated rightly according to the afore-method. The results indicated that the PDV model could be used to discriminate the genotypes with different degrees of difference. Eventually, six genotypes were utilized to study the expert system based on their different total number of core repeat units. They were discriminated rightly. The results provide the possibility for establishment of expert system for other STR loci.The paper has revised the pharmacopoeial standard of Oleum Viticis Negundo. It provides technical support for the revision of Ch. P (2010) and makes the clinical use of Oleum Viticis Negundo safer and more effective. The expert systems for STR genotyping were established making the determination of STR genotypes simple, rapid and low cost.
