Meanwhile 32 cases of normal gastric tissue were chosen as control. E-cadherin, NF-kb and Snail were detected by immunohistrochemistry staining. Kaplan-Meier survival analysis was performed for 101 cases with survival time. We analyzed the prognostic factors by Cox regression model in these cases.Results:1,The positive rates for E-cadherin of the gastric carcinoma, paracancer and control were respectively 22.2%(42/189),55.6%(30/54) and 100%(32/32). The positive rate for E-cadherin of the gastric carcinoma were lower than that of the paracancer as well as the control (P<0.001). The positives rates for E-cadherin of the differentiated gastric carcinoma and undifferentiated gastric carcinoma were 33.7% (30/89) and 11.0%(11/100) respectively. There was significant difference between the two groups(P=0.002). The positive rates for E-cadherin of the gastric carcinoma with invasion under serosa and that with invasion out of serosa were respectively 37.0%(17/46) and 16.8%(24/143), the former was higher than the latter(P =0.003).The result of Spearman correlation analysis was that the expression of E-cadherin had negative correlation with the depth of invasion (r=-0.209, P=0.004). The positive rate for E-cadherin of intestinal gastric carcinoma was higher than that of diffused gastric carcinoma(P=0.001)..2. The positive rates for NF-kB of the gastric carcinoma, paracancer and control were 75.1%(142/189),42.6%(23/54) and 15.6%(5/32) respectively, with significant difference among the three case groups(P<0.05). The positive rates for NF-kB of the gastric carcinoma with lymph node metastasis and that of those without lymph node metastasis were 81.0%(94/116) and 65.6%(48/73) respectively, the former was higher than the latter (χ2=5.600, P=0.018). The positive rates for NF-kB of the gastric carcinoma with invasion under serosa and that with invasion out of serosa were 60.9%(28/46) and 79.7%(114/143) respectively, There was significant difference between the two groups (χ2=6.619,P=0.010).The positives rates for NF-kB of the differentiated gastric carcinoma and undifferentiated gastric carcinoma were 67.4%(60/89) and 82.0%(82/100) respectively. There was significant difference between the two groups(χ2=5.361,,P=0.021). The positive rate for NF-kB of diffused gastric carcinoma was higher than that of intestinal gastric carcinoma (χ2 =7.284, P=0.007).3. The positive rates for Snail of the gastric carcinoma, paracancer and control were 84.13%(159/189),48.45%(26/54) and 18.75%(6/32) respectively. The positive rate for Snail of the gastric carcinoma were higher than that of the paracancer as well as the control (P<0.001) The positives rates for Snail of the differentiated gastric carcinoma and undifferentiated gastric carcinoma were 68.5%(61/89) and 82.0% (82/100) respectively. There was significant difference between the two groups (χ2 =4.586, P=0.032). The positive rates for Snail of the gastric carcinoma with lymph node metastasis and that of those without lymph node metastasis were 81.0% (94/116) and 67.1%(49/73) respectively, the former was higher than the latter (χ2 =4.708, P=0.03). The positive rates for Snail of the gastric carcinoma with invasion under serosa and that with invasion out of serosa were 63.0%(29/46) and 79.7% (114/143) respectively, There was significant difference between the two groups(χ2 =5.549, P=0.018). The positive rate for Snail of diffused gastric carcinoma was higher than that of intestinal gastric carcinoma (χ2=8.563, P=0.003)4.101 cases with survival time were performed by Log-rank test of Kaplan-Meier survival analysis. The survival time of patients with negative Snail, negative NF-kB and positive E-cadherin was higher than those of patients with positive Snail, positive NF-kB and negative E-cadherin (P<0.05) respectively. The survival time of patients without lymph node metastasis was higher than that with lymph node metastasis(χ2= 19.634, P=0.000). The survival time of patients with invasion under serosa was higher than that out of serosa(χ2=20.279, P=0.000). The survival time of patients with differentiated gastric carcinoma was higher than that with undifferentiated gastric carcinoma(χ2=4.561, P=0.033). The survival time of patients whose diameter is smaller than 5cm (Φ<5cm) was higher that of patients whose diameter is bigger than 5cm(Φ≥5cm) (χ2=4.336, P=0.037).5. E-cadherin, NF-kB,Snail, depth of invasion, lymph node metastasis were independent prognostic factors by COX regression analysis.Conclusions:1. The expression of Snail and NF-kB was increased in gastric carcinoma, meanwhile E-cadherin was reduced. These indicated that E-cadherin NF-kB and Snail contributed to the process of the genesis and progress of gastric carcinoma.2. The reduced E-cadherin as well as the increased NF-kb and Snail maybe was the early incidents of gastric cancer.3. Such factors as E-cadherin, NF-kB, Snail, tumor size, differentiation, lymph node metastasis, depth of invasion were closely associated with survival time of patient with gastric cancer.
