| TB remains a major global threat to human health. There are almost 17-20 million people infected with MTB (Mycobacterium tuberculosis) worldwidely, and 3 million new patients annually. China ranks top after India as heavy-burden TB incidence. Latent MTB infection directly led to a long course of TB treatment, poor efficacy, and easy to relapse after chemotherapy, because of course time turns so long, most people can not afford the necessary medical costs, patients are not fully chemotherapy, and this is the reasons of high relapse rate and drug resistance of TB cases increased, also that is why TB is hard to cure.The maintenance of genome stability is the crux for organism survival. Base excision repair (BER) is a major DNA repair pathway. This is particularly important for Mycobacterium tuberculosis and other intracellular pathogens, fpg is the key enzyme in base excision repair. Mycobaterium genome comparison revealed that pathogenic M.tuberculosis harbor more genes involved in the BER. This might implicate that BER is closely related to the intracellular survival and pathogenesis of this bug. This justifies the BER pathway and crucial enzymes as promising targets for novel antibiotics.fpg of M. tuberculosis H37Rv was cloned into pET32a(+) and expressed in E.coli BL21(DE3) with the induction of IPTQ and subjected to Ni2+ affinity chromatograph. The comparison of the control and fpg transformed host demonstrated that the fpg gene plays a role in the peroxide hydrogen and UV irradiation resistance. |
