Study On Quanititative Analyses Of Chemical Composition Of Naotai Prescription Decoction And Capsules

Objective:NaoTai Prescription, which is made from Radix Astragali,Rhizoma Chuanxiong,Pheretima and Batrgticated Silkworms, is an effective prescription intreating cere infarction。In order to understand its materials of treatment and ensure the quality of capsules with modern technology production and improve technology, The auther has made a study on its decoction and capsules preliminary quanititative analyses of three kinds of major chemical compositions(Semifinished protein,Free amino acid,Polysaccharide) and three understand active single components(Ferulic acid,Ligustrazine,AstragalosideⅣ)。Methods:To decoction and capsules, determination of dry extract with normal atmosphere and vacuum drier,the contents of Semifinished protein,Free amino acid,Polysaccharide were determined by semimicro-method and colorimetric analysis, the contents of Ferulic acid,Ligustrazine,Astragaloside IV were determined by R-HPLC。Assay method by experimental study was validated in the research of their content。Results:By experimental study, the rate of getting dry extract with vacuum drier:the number of decoction was 26.2%, the number of capsules was 29.1%, the toal solid quanity from plants is 1.6 times from herbs;the rate of getting soaking materials(g/g):the number of Semifinished protein was 13.1% in decoction and 14.7% in capsules, the number of Free amino acid was 3.11% in decoction and 3.20% in capsules, the number of Polysaccharide was 9.5% in decoction and 12.0% in capsules, the number of Oxalic acid ammonium was 0.631% in decoction and 0.0643% in capsules.Thus it can seen, the content of Oxalic acid ammonium and Polysaccharide was great difference, the number of Semifinished protein,Polysaccharide and Oxalic acid ammonium in dry extract of capsules was 50.5%,41.1% and 2.81% and their sum was 93.8%; the number of Semifinished protein,Polysaccharide and Oxalic acid ammonium in solute of decoction was 50.8%,36.3% and 8.50% and their sum was 94.9%, consequently, they are major chemical composition.By the experimental study on understand active single components, the content of soaking materials (mg/100g), the content of Ferulic acid was 17.5mg in decoction and 8.75mg in capsules, the content of Ligustrazine was 8.42mg in decoction and 1.89mg in capsules, the content of Astragaloside IV was 42.8mg in decoction and 45.8mg in capsules。We knew that the Ligustrazine and Ferulic acid from capsules had obviously lose and the quanity of astragaloside IV from capsules is higher than from decoction。...
