Objectives to investigate the mechanism of Vascular endothelial growth factors regulates epithelial-to-mesenchymal transition by Snail signaling pathway in gastric cancer. Methods The expression of VEGF,Snail,E-cadherin and Vimentin proteins in 69 cases of gastric cancer tissues were detected by immunohistochemical staining, analysis the correlation between Snail,E-cadherin,Vimentin and VEGF. Human poorly differentiated gastric cancer cell line MGC-803 were cultured with different concentrations of VEGF and some concentration VEGF antibody. Detect the expression of VEGF,Snail,E-cadherin and Vimentin mRNA by RT-PCR. Results 69 cases of gastric cancer, the positive rate of VEGF was 73.9%(51/69); Snail was 78.2% (54/69); E-cadherin was 36.2%(25/69);Vimentin was 49.3%(34/59). The expression of VEGF,Snail,E-cadherin and Vimentin proteins was closely related to the depth of tumor invasion,differentiation,lymph node metastasis and TNM staging(P<0.05), and was not related to gender,age,tumor location and tumer size(P>0.05).The expression of Snail mRNA and Vimentin mRNA was significantly increased when the VEGF concentration cultured MGC-803 from 0.1 to 0.5ng/ml, and the expression was declined slightly when the VEGF concentration from 0.5 to 5ng/ml.The expression of E-cadherin mRNA was decreased when the VEGF concentration from0.1,0.5,5ng/ml changes. The expression of Snail mRNA and Vimentin mRNA was decreased and E-cadherin mRNA was increased when the MGC-803 was cultured with 5ug/ml VEGF antibody.Conclusions VEGF promotes invasive and metastasis via epithelial-to-mesenchymal transition ,VEGF-Snail signaling pathway is the possible mechanism.
