Cynanchum komarovii Al. Iljinski (CK) is a kind of Chinese herb. CK is usually used as insecticide by local villagers. In Chinese traditional treatment, however, CK can relieve pain, remove fever,check diarrhea and prevent cholecystitis. As reported in recent 20 years, CK has the potential of anti-asthma and anti-inflammation, and the potent compounds in CK are mainly alkaloids (ACKs).But the mechanism of anti-inflammation by ACKs has not been manifested yet, so we try to investigate it through this study.PartⅠEffects of ACKs on rat carrageenin-induced pleurisyObjective To study the effects of alkaloids from Cynanchum komarovii Al. Iljinski on rat carrageenin-induced pleurisy.Methods We pretreated the rats with indomethacin (10mg/kg) or ACKs (3, 6, 12mg/kg) by intragastric administration and intraperitoneal injection, respectively, and stimulated them with carrageenin (2mg/rat) by intrapleural injection 30min later to make pleurisy models. After 4h, we sacrificed the rats and collected the pleural exudate and lung to detect the following parameters such as exudate volume, leukocyte number and observed the lung injury by light microscopy.Results Pretreatment of rats with ACKs inhibited dose-dependently carrageenin-induced pleurisy. They reduced pleural exudate formation and leukocyte migration. And the lung injury was attenuated obviously. Conclusion ACKs has an obvious anti-inflammatory effect on rat carrageenin-induced pleurisy.PartⅡInfluence of ACKs on the cytokines in the exudate produced by rat carrageenin-induced pleurisyObjective To study the influence of ACKs on the cytokines in the exudate produced by rat carrageenin-induced pleurisy.Methods We pretreated the rats with indomethacin (10mg/kg) or ACKs (3, 6, 12mg/kg) by intragastric administration and intraperitoneal injection, respectively, and stimulated them with carrageenin (2mg/rat) by intrapleural injection 30min later to make pleurisy models. PartⅢInfluence of ACKs on NO and iNOS produced by rat carrageenin-induced pleurisyObjective To study the influence of ACKs on NO and iNOS produced by rat carrageenin-induced pleurisy.Methods We pretreated the rats with indomethacin (10mg/kg) or ACKs (3, 6, 12mg/kg) by intragastric administration and intraperitoneal injection, respectively, and stimulated them with carrageenin (2mg/rat) by intrapleural injection 30min later to make pleurisy models. After 4h, we sacrificed the rats and collected the pleural exudate and the lung to detect the levels of NO and iNOS respectively.Results Pretreatment of rats with ACKs reduced the level of NO in the exudate produced by rat carrageenin-induced pleurisy and the level of iNOS in the lung.Conclusion ACKs inhibits rat carrageenin-induced pleurisy by reducing the production of NO through the inhibition of the activity of iNOS.PartⅣInfluence of ACKs on the oxidative injury of lung in rat carrageenin-induced pleurisyObjective To study the influence of ACKs on the anti-oxide activity in rat carrageenin-induced pleurisy.Methods We pretreated the rats with indomethacin (10mg/kg) or ACKs (3, 6, 12mg/kg) by intragastric administration and intraperitoneal injection, respectively, and stimulated them with carrageenin (2mg/rat) by intrapleural injection 30min later to make pleurisy models. After 4h, we sacrificed the rats and collected the lung to detect the levels of MDA and SOD.Results Pretreatment of rats with ACKs reduced the level of MDA in the lung but raised the level of SOD.Conclusion ACKs inhibits rat carrageenin-induced pleurisy by increasing the activity of anti-oxide in vivo.
