| Objective: Detection of expression level ofβ2AR,β-arrestin2, NF-κBP65,STAT6 before and after medical treatment of Ulcerative Colitis in rats .Method: SD rats were randomly divided into four groups: control group, model group, Mesalazine western medicine group and wumeiwan traditional Chinese medicine group,8 rats for each group.After the model is successful established, the rats of the control groups and model groups with normal saline 3ml gavage, western medicine group with the slurry of 50mg/ml Mesalazine 50mg/100g gavage, traditional Chinese medicine group was given 0.515g/ml of Wu Mei Wan liquid 3ml ig, each group were continuously administered 15 days. Tissues of spleen and colon were obtained and detected the level of expression ofβ2AR,β-arrestin2, NF-κBP65,STAT6 using western blot method and immunohistochemistryResult:β2AR,β-arrestin2 expression increased more in model group than in control group(P<0.01), and after the treatment of WuMeiWan and Mesalazine, the expression of both are decreased significantly(P<0.01), and the difference between the two groups have no statistically significant. (P>0.05) and NF-κBP65 and STAT6 less express in normal tissues(P<0.01), the expression of NF-κBP65 and STAT6 significantly increased by the drug treatment group, (P<0.01) including WuMeiWan and Mesalazine than the model group. The difference between the two groups has no statistically significant. (P>0.05)Conclusion: One of the mechanisms of Wu Mei Wan to treat ulcerative colitis is achieved by regulating theβ2AR-β-arrestin2- NF-κBp65/STAT6 signal transduction pathway. |
