Objectives: To study the preventive effects of Lentinula Capsule on hyperlipidemia by observing the influence of Lentinula Capsule on lipid metabolism, oxidative stress and liver metabolism.Methods: 50 male SD rats were randomly divided into 5 groups, 10 in each, according to body weight and fasting serum total cholesterol concentration after a week of chow feeding. They were one control group on chow, one model group fed with high-fat diet and three Lentinula Capsule groups fed with high-fat diet and with low, medium and high dose of Lentinula Capsule per day respectively. During 28 days of experimental period, the Lentinula Capsule groups were adiministrated 0.4 g/kg, 1.2 g/kg and 2.4 g/kg of Lentinula Capsule solution orally, whereas the rats of model group high-fat and chow -fed control group were gave the same volume saline. Food intake and weight gain were measured weekly. At the beginning of the experiment, tail vein blood were taken to separate serum after the rats were fasted 12 hours orernight. At the end of the experiment, the rats were killed by decapitation after overnight fasting. Blood samples were collected and livers removed and weighed. Serum TC, TG and HDL-C concentrations were measured. Serum MDA content, SOD and GSH-PX activity were determined. All statistical analyses were conducted using packages SPSS13.0. All data are presented as means±SD ( x_±s).Resu1ts: (l) The effect of Lentinula Capsule on body weight of rats: At the beginning of the experiment, there were no significant difference in body weight among groups. The average weight of each group increased significantly with time, while no significant differences was found among groups at corresponding time points(P>0.05). (2) Effects of Lentinula Capsule on total food intake and total energy intake in experimental rats: Groups administered with different doses of Lentinula Capsule and model group of high-fat all reduced their total food intake significantly in comparison to chow-fed control group. Rats with high-fat diet had more energy intake than chow-fed control and high fat feeding rats with Lentinula Capsule administration showed reduced energy intake.⑶The impact of Lentinula Capsule on liver metabolism: By observing external appearance of livers, we found the livers of chow-fed control groups were in the normal state, model group of high fat diet showed severely pathological state, the groups treated with Lentinula Capsule had less severe liver injury than model group of high fat diet as judged by gross liver appearance. The liver weight and percent body weight of liver in model group of high fat diet were higher than chow-fed control group and Lentinula Capsule groups (P <0.05). (4) Effects of Lentinula Capsule in the regulation of blood lipids: Serum TC, TG, LDL-C concentrations and AI were increased and HDL-C concentration were decreased significantly in model group of high fat diet compared with chow-fed control group(P <0.05). While compared with model group of high fat diet, Lentinula Capsule groups decreased TC, LDL-C concentrations and AI and increased HDL-C concentration. Lentinula Capsule had not influence on TG in this experiment. (5) Effects of Lentinula Capsule in anti-lipid peroxidation: Compared with the chow-fed control group, serum MDA concentration were increased and SOD and GSH-PX activity were decreased in model group of high fat diet. In comparison to model group of high fat diet, Lentinula Capsule effectively reduced serum MDA concentration, and increased the SOD and the GSH-PX activity, with significantly different (P<0.05 ).Conc1us1ons: The findings showed that Lentinula Capsule reduced significantly the food intake of rats with hyperlipidemia so that their energy intake also were decreased. Lentinula Capsule had a positive effect on regulation of blood lipids,anti-lipid peroxidation and regulation of liver metabolism for the prevention of hyperlipidemia, atherosclerosis, fatty liver,as well as subsequent development of coronary heart disease and myocardial infarction and so on.
