| ObjectiveTo detect the expression of IL-24 and VEGF-C in normal cervix,cervical intraepithelial neoplasia(CINâ…¡-â…¢) and invasive cervical sqamous cell carcinoma(CSC).And to assess the relationship of both in the clinicopathological parameters and lymphatic metastasis of CSC.Materials and Methods83 case of Patients were treated between October 2007 and March 2011 were enrolled in this study.There were 10 cases of normal cervix,12 cases of cervical intraepithelial neoplasia(CIN) and 61 cases of invasive cervical sqamous cell carcinoma(CSC) in them, international Federation of Gynecology and Obstetrics(FIGO) stage were 7 as FIGO stage IA2,20 as FIGO stage IB 1,10 as FIGO stage IB2,21 as FIGO stage IIA and 3 as FIGO stageâ…¡B.29 cases were lymphatic metastasis that confirmed by histological review and 32 were negaive. Based on the analysis of histological grade,6 cases were HG1,31 were HG2,and 24 were HG3.Immunohistochemical(IHC) staining with the antibidies against IL-24 and VEGF-C was used to examine the expression of them in normal cervix,cervical intraepithelial neoplasia(CINâ…¡-â…¢) and invasive cervical sqamous cell carcinoma(CSC).All statistcal calculations were performed using SPSS 15.0. Result1.The expression of IL-24 in normal cervix,cervical intraepithelial neoplasia(CINâ…¡-â…¢) and invasive cervical sqamous cell carcinoma(CSC)IL-24 were mostly localized in the cytoplasm of the normal squamous cells and the positively stained cells showed a brown-yellow color.The positive rates were 100.00%(10/10),75.00%(9/12) and 32.79%(20/61) in normal cervix,CINâ…¡-â…¢and invasive CSC samples,respectively. There were significant differences of positive expressive rates of IL-24 among normal cervix,CINâ…¡-â…¢and invasive CSC samples(x2=20.001, P=0.000).There was no significant differences in the two-two comparisons between the normal cervix and CINâ…¡-â…¢(x1=1.161, P=0.281). The rest were significant differences each other (all P=0.000).2.Association of the expression of IL-24 with clinical and pathological parameters in invasive CSCThe expression levels of IL-24 decreased with the progress of Its FIGO stage in invasive CSC, but not with age,histological grade and depth of stromal invasion(all P>0.05). The expression levels of IL-24 was lower in invasive CSC lymph node metastasisis than that with without lymph node metastasis (P<0.05).3. The expression of VEGF-C in normal cervix,cervical intraepithelial neoplasia(CINâ…¡-â…¢) and invasive cervical sqamous cell carcinoma(CSC)VEGF-C were mostly localized in the cytoplasm of the cancer cell in the examined CSC samples and the positively stained cells showed a brown-yellow color.The positive rates were 0.00%(0/10),16.67%(2/12) and 54.10%(33/61) in normal cervix,CINâ…¡-â…¢and invasive CSC samples,respectively. There were significant differences of positive expressive rates of VEGF-C among normal cervix,CINâ…¡-â…¢and invasive CSC samples(X2=13.834, P=0.000).There was no significant differences in the two-two comparisons between the normal cervix and CINâ…¡-â…¢(x2=0.371, P=0.542). The rest were significant differences each other (all P<0.05).4.Association of the expression of VEGF-C with clinical and pathological parameters in invasive CSCThe expression levels of VEGF-C was higher in invasive CSC with lymph node metastasis is than that without lymph node metastasis (P<0.05),but not with age, FIGO stage,histological grade and depth of stromal invasion(all P>0.05).5.Correlation of expression of IL-24 and VEGF-C in invasive CSCThe expression levels of IL-24 were negatively correlated with VEGF-C(rs=0.433, P=0.000).Conclusion 1. The expression levels of IL-24 were gradually decreased among normal cervix,CINâ…¡-â…¢and invasive CSC samples,but not with age,histological grade and depth of stromal invasion.The expression levels of IL-24 decreased with the progress of Its FIGO stage, and the low expression rates was closely related to lymph node metastasis.This demonstrates that the expression of IL-24 maybe corrlates with the tumorigensis of CSC in the lymphatic metastasis.2. The expression levels of VEGF-C were gradually increased among normal cervix,CINâ…¡-â…¢and invasive CSC samples.Its high expression rates was closely related to lymph node metastasis,but not with age,FIGO stage,histological grade and depth of stromal invasion.This demonstrates that the expression of VEGF-C maybe corrlates with the development of CSC3.
