[Objective]To study the influence and clinical significance of nucleoside analogs antiviral therapy on the level of Thl/Th2 cytokines in serum of hepatitis B virus-related cirrhosis, and the changes of the level of cytokines and its relationship with HBVDNA capacity, whether these can be used as curative effect forecasting standard and curative effect evaluation criteria for nucleoside analogs antiviral therapy, to discuss the significance of applying immunomodulater or exogenous cytokine therapy to chronic hepatitis B.[Methods]1,Sources of cases:(1) antiviral group:25 cases with hepatitis B virus-related cirrhosis accept nucleoside analogs antiviral therapy and ordinary liver protecting treatment, Matched group:10 cases with hepatitis B virus-related cirrhosis only accept ordinary liver protecting treatment.2, Test method:(1) To apply double-antibody sandwich ELISA to test the level of serum IFN-γ,IL-4,IL-6,IL-8,IL-18, (2) To apply Real-time fluorescent quantitative PCR to test HBVDNA. (3) To test patients'liver function in different groups with automatic biochemistry analyzer.[Results]A significant difference can be reflected by the level of IL-4,IL-6,IL-8,IL-18,IFN-γ,ALT,ALB,TBIL,HBVDNA before and after the therapy of nucleoside analogs antiviral((P<0.05),and relevance can be also seen between the level of cytokines IL-6,IFN-γand the level of HBVDNA (P<0.05). There was a significant difference in the level of IL-8,ALT,ALB before and after the ordinary liver protecting treatment for the patients in matched group, while the level of IL-4,IL-6,IL-18,IFN-γ,TBiL,HBVDNA had not(P>0.05);in the group with nucleoside analogs antiviral therapy, no significant difference can be found in the level of cytokines in different classification of liver function(P> 0.05),while there was a significant difference of the level of IL-4,IL-6,IFN-y before and after the different nucleoside analogs antiviral therapy (P<0.05)[Conclusions]1,Nucleoside analogs not only suppress HBVDNA for hepatitis B virus-related cirrhosis, but also reduce the content of IL-4,IL-6,IL-8 obviously, increase the content of IL-18,IFN-γ, therefore modulate the balance of Thl/Th2 cytokine network.2,The content of IL-6 is positively associated with HBVDNA, while there is a negative relation between the content of IFN-γand HBVDNA.
